摘  要:小鹅瘟卵黄抗体(IgY)可通过被动免疫预防小鹅瘟病,脂类存在直接影响其预防效果,甚至引起发病。文章在制备小鹅瘟卵黄抗体过程中,应用酸化水脱脂,探究酸化水稀释度、pH、作用时间及离心力对除脂率及IgY提取的影响,筛选除脂率、琼扩效价及蛋白浓度最高的试验参数组合,确定优化生产工艺。结果表明,酸化水稀释度、pH、作用时间、离心力对除脂率影响显著,优化参数组合为:酸化水1:8稀释、pH 5.2、4℃存放15 h(过夜)、1 900 g离心20 min工艺条件下,IgY含量与琼扩效价均高于其他试验组,脱脂效果显著,适用于小鹅瘟IgY脱脂工艺,为工业规模化生产提供参考。Gosling yolk antibody(IgY) could prevent gosling plague by passive immunity. Lipids in gosling plague IgY could directly affect the preventive effect, and even led to diseases. In this experiment,acidi ed water degreasing was used in the process of preparing gosl yolk antibody, and the effects of acidified water dilution titer, pH, time of operation and centrifugal force were explored on degreasing rate and IgY extraction, then experimental combinations of degreasing rate, agar expansion valence, and protein concentration were selected to determine the optimal production process. The results showed that the acidi ed water dilution titer, pH, duration, centrifugal force had significant effects on degreasing rate. Under the optimum procedure, acidified water dilution titer 1:8, pH 5.2, 4 ℃ 15 h(overnight), 1 900 g centrifugal 20 min, IgY content and agar expansion valence were higher than those of other combinations, with remarkable degreasing effect.This approach was suitable to gosling plague IgY degreasing process, and provided the optimal technological conditions for industrial scale production.
