摘 要:目的观察健脾柔肝息风汤对抽动障碍(tic disorder,TD)模型大鼠脑组织中多巴胺和高香草分组干预后酸(homovanillic acid, HVA)含量的影响,探讨其作用机制。方法建立空白组,模型组,氟哌啶醇组,健脾柔肝息风汤高、中、低剂量组共6组,通过腹腔注射亚氨基二丙腈,建立抽动障碍模型大鼠,分组干预后再采用酶联免疫法(ELISA)测定脑组织中多巴胺(dopamine, DA)和高香草酸(homovanillic acid, HVA)的含量。结果与空白组比较,模型对照组DA及HVA含量均降低,差异有统计学意义(P<0.01);与模型组比较,健脾柔肝息风汤高、中、低剂量组及氟哌啶醇组DA与HVA含量均升高(P<0.05或P<0.01)。结论 DA能系统及代谢产物HVA平衡失调与抽动障碍发病有关,而健脾柔肝息风汤具有调节DA能系统及代谢产物平衡的作用。Objective To observe the effect of Jianpi Rougan Xifeng Decoction on the content of dopamine(DA)and homovanillic acid(HVA)in the brain tissue of model rats with tic disorder,and to explore its mechanism.Methods The rats were divided into six groups,i.e.,blank group,model group,haloperidol group,and Jianpi Rougan Xifeng Decoction groups(high,medium,and low doses).A rat model of tic disorder was established by intraperitoneal injection of iminodipropionitrile.After administration of corresponding treatment for each group,the content of DA and HVA in the brain tissue was determined by enzyme-linked immunosorbent assay.Results Compared with the blank group,the model group had significantly reduced content of DA and HVA(P<0.01);the content of DA and HVA was significantly higher in the Jianpi Rougan Xifeng Decoction groups(high,medium,and low doses)and haloperidol group than in the model group(P<0.05or P<0.01).Conclusion The imbalance between the dopaminergic system and its metabolite(HVA)is related to the development of tic disorder,while Jianpi Rougan Xifeng Decoction can regulate the balance between the dopaminergic system and its metabolite.
