摘  要:目的通过行为绝望模型—悬尾实验(TST)和不可预测性慢性温和应激(CUMS),探讨刺蒺藜总皂苷对小鼠抑郁样行为的作用以及对神经递质的影响。方法 (1)TST中连续给予刺蒺藜总皂苷10 d后,通过测定悬尾不动时间、开场实验(OFT)测定5 min内走过的路程,观察其兴奋作用;(2)CUMS实验,连续给予刺蒺藜总皂苷15d,通过强迫游泳实验,评价刺蒺藜总皂苷的抗抑郁活性。(3)机制探讨:通过测定5-羟色胺(5-HT)、去甲肾上腺素(NE)、脑源性神经营养因子(BDNF)的水平,探讨刺蒺藜总皂苷的抗抑郁作用机制。结果行为绝望模型—悬尾实验中,与空白组相比,刺蒺藜总皂苷可以显著降低小鼠的不动时间、提高小鼠OFT的自主活动(均P<0.05)。CUMS实验中,与模型组比,刺蒺藜总皂苷可以显著降低强迫游泳中的不动时间(均P<0.05)。此外,刺蒺藜总皂苷可显著增加小鼠血清中5-HT的水平(P<0.05),对NE、海马中BDNF含量也有影响。结论刺蒺藜总皂苷对小鼠有一定的抗抑郁作用,作用机制可能与提高5-HT的水平有关。Objective The effects of total saponins of Tribulus terrestris L on depressive behavior and neurotransmitters in mice were investigated by behavioral despair model-tail suspension test(TST)and chronic unpredictable mild stress(CUMS).Methods①In the tail suspension experiment,after 10 days of continuous administration of total saponins of Tribulus terrestris L,the time of suspension experiment was determined,and the distance in 5 min was measured by open field test(OFT),and the excitatory effect was observed.②CUMS test was used to evaluate the antidepressant activity of total saponins of Tribulus terrestris L by forced swimming after 15 days of continuous administration.③The concentration of 5-HT and NE in serum,the concentration of BDNF in hippocampus as indexes were measured to estimate the mechanism of antidepressant action.Results The behavioral despair mice model-TST showed that total saponins of Tribulus terrestris L could significantly reduce the immobile time of the mice and improve the spontaneous activity of the opening experiment of mice(all P<0.05)compared with the blank control group.The CUMS show that total saponins of Tribulus terrestris L can significantly reduce the immobilization time in the forced swimming test(all P<0.05)compared with CUMS model group.In addition,the concentration of 5-HT and NE in serum can be significantly increased(P<0.05)after receiving total saponins of Tribulus terrestris L.Total saponins of Tribulus terrestris L have effect on the concentration of 5-HT and NE in serum and the concentration of BDNF in hippocampus,too.Conclusion Total saponins of Tribulus terrestris L have certain antidepressant activity in mice.The mechanism of action may be related to increasing the level of 5-HT.
