摘　　要：目的:探讨白细胞介素17A(IL-17A)对RAW264. 7巨噬细胞三磷酸腺苷结合盒转运体A1(ABCA1)表达的影响。方法:用不同浓度及的IL-17A处理小鼠RAW264. 7细胞6 h或24 h,或用同一浓度IL-17A处理RAW264. 7细胞不同时间;采用RT-qPCR及Western blot法检测细胞ABCA1的mRNA和蛋白表达情况;采用NBD-胆固醇法检测细胞胆固醇流出情况;采用油红O染色法测定细胞脂质蓄积情况。结果:与对照组相比,IL-17A可以增加RAW264. 7巨噬细胞ABCA1蛋白的表达水平,但不影响ABCA1 mRNA的表达。经IL-17A干预,RAW264. 7巨噬细胞内胆固醇向Apo A-1流出增多,同时细胞脂质蓄积显著减少(P <0. 01)。结论:IL-17A可以增加RAW264. 7巨噬细胞ABCA1的表达水平,其机制并不是通过转录水平实现的,这种影响可能与其抗动脉粥样硬化作用有关。AIM:To investigate the effects of interleukin-17A(IL-17A)on the expression of adenosine triphosphate binding cassette transporter A1(ABCA1)in RAW264.7macrophages.METHODS:Mouse RAW264.7macrophages were treated with IL-17A at different concentrations for6or24h,or treated with IL-17A at the same concentration for different time.The expression of ABCA1at mRNA and protein levels was determined by RT-qPCR and Western blot,respectively.Cholesterol efflux to apolipoprotein A1(ApoA-1)was evaluated by NBD-cholesterol method.Lipid accumulation in the cells was evaluated by Oil Red O staining.RESULTS:Compared with control group,IL-17A increased the expression of ABCA1at protein level in the RAW264.7cells significantly(P<0.05),but had no effect on the mRNA expression of ABCA1.In addition,cholesterol efflux to ApoA-1was increased and lipid accumulation in the RAW264.7cells was decreased obviously after treatment with IL-17A.CONCLUSION:IL-17A increases the protein expression of ABCA1but not at mRNA level in the RAW264.7macrophages,which may be correlated with its anti-atherosclerosis effect.
