高效液相色谱法测定追风伞提取物中5种酚酸类成分含量

摘　　要：目的建立同时测定追风伞提取物中绿原酸、芦丁、异鼠李素-3-O-β-D-葡萄糖苷、木犀草素及刺槐素含量的高效液相色谱(HPLC)法。方法色谱柱为Phenomenex C18柱(250 mm×4. 6 mm,5μm),流动相为甲醇(A)-0. 2%醋酸水溶液(B),梯度洗脱,流速为1 m L/min,柱温为30℃,检测波长为334 nm。结果绿原酸、芦丁、异鼠李素-3-O-β-D-葡萄糖苷、木犀草素及刺槐素质量浓度分别在4. 132 5～165. 300 0 mg/L,6. 440 0～257. 600 0 mg/L,3. 980 0～159. 200 0 mg/L,4. 352 5～174. 100 0 mg/L,3. 592 5～143. 700 0 mg/L范围内与峰面积线性关系良好,平均回收率分别为98. 28%,99. 10%,98. 23%,98. 61%,98. 02%,其样品含量分别为1. 701,3. 940,2. 179,3. 070,1. 881 mg/g。结论该方法快速、准确、重复性好,可用于评价和控制追风伞提取物的质量。Objective To establish an HPLC method for content determination of chlorogenic acid,rutin,isorhamnetin-3-O-β-D-glucoside,luteolin and acacetin in the extract of Lysimachia paridiformis. Methods The chromatographic column was Phenomenex C18 column( 250 mm × 4. 6 mm,5 μm),the mobile phase was methanol( A)-0. 2% acetic acid( B),gradient elution,the flow rate was 1 m L/min,the column temperature was 30 ℃,and the detection wavelength was 334 nm. Results Good linear relationship was found in the range of 4. 132 5-165. 300 0 mg/L for chlorogenic acid,6. 440 0-257. 600 0 mg/L for rutin,3. 980 0-159. 200 0 mg/L for isorhamnetin-3-O-β-D-glucoside,4. 352 5-174. 100 0 mg/L for luteolin,3. 592 5-143. 700 0 mg/L for acacetin. The average recovery rates for chlorogenic acid,rutin,isorhamnetin-3-O-β-D-glucoside,luteolin and acacetin were 98. 28%,99. 10%,98. 23%,98. 61 % and 98. 02 %,respectively,and the content of chlorogenic acid,rutin,isorhamnetin-3-O-β-D-glucoside,luteolin and acacetin in the extract of Lysimachia paridiformis were 1. 701,3. 940,2. 179,3. 070,1. 881 mg/g,respectively. Conclusion The method is rapid,accurate and reproducible,which can be used for evaluating and controlling the quality standard of the extract of Lysimachia paridiformis.
