摘 要:目的:观察糖尿病大血管纤维化大鼠模型中假性蛋白激酶3(TRIB3)及Toll样受体4(TLR4)的表达情况。方法:雄性SD大鼠200只,随机选30只为对照组;余170只予高脂高糖饲料喂养,腹腔注射链脲佐菌素(STZ)、高脂高糖饲料持续喂养,进行糖尿病血管纤维化造模,造模成功后选取30只为模型组,30只为桃仁干预组。前者予生理盐水10 mL·kg^(-1)·d^(-1)灌胃,后者予桃仁颗粒剂水溶液10 mL·kg^(-1)·d^(-1)灌胃连续7周。7周后三组分别取材,保存标本。免疫组化法检测TLR4沉积部位,Western blotting检测TLR4表达情况,实时荧光定量PCR技术检测TRIB3基因的表达情况。结果:免疫组化检测分析,对照组及桃仁干预组大鼠血管内皮细胞及中膜平滑肌细胞中有散在黄色物质,呈弱阳性改变;模型组可见大量棕黄色物质表达呈强阳性改变。TLR4的Western blotting检测,桃仁干预组表达(0.5513±0.0432)较对照组(0.4021±0.0449)高,差异有统计学意义(P<0.05);与模型组(1.8342±0.0342)比较,表达明显减低,差异具有统计学意义(P<0.01);模型组与对照组比较,TLR4表达明显增高,差异有统计学意义(P<0.01)。实时荧光定量PCR技术检测TRIB3基因在桃仁干预组(1.3638±0.0948)较模型组(2.3241±0.0654)表达低,差异具有统计学意义(P<0.01);桃仁干预组较对照组(1.0000±0.0000)表达高,差异具有统计学意义(P<0.05);模型组较对照组表达明显升高,差异具有统计学意义(P<0.01)。结论:糖尿病大血管纤维化大鼠股动脉中TRIB3基因和TLR4蛋白出现高表达,中药桃仁可以降低其表达。Objective To observe the expression of tribbles3gene(TRIB3)and Toll-like receptor4(TLR4)in diabetic macrovascular fibrosis rat model.Methods Two hundred male SD rats were used as the seudy.Thirty male SD rats were randomly selected as control group,and the remaining170rats were established diabetic vascular fibrosis model by feeding with high-fat and high-sugar diet,intraperitoneal injection of streptozotocin(STZ),and high-fat and high-sugar diet for sequentially continuous.After successful modeling,30rats were selected as model group,30as peach kernel intervention group.The former was intragastrically administered with10mL·kg-1·d-1saline,while the latter was intragastrically administered with10mL·kg-1·d-1peach kernel granule solution for7weeks.After7weeks,the specimens were collected and preserved in three groups.Immunohistochemical method was used to detect the location of TLR4deposition.Western blotting was used to detect the expression of TLR4,and real-time fluorescence quantitative PCR was used to detect the expression of TRIB3.Results Immunohistochemical analysis showed that there were scattered yellow substances in vascular endothelial cells and smooth muscle cells in the control group and peach kernel intervention group,showing weak positive changes,and a large number of brown substances in the model group showed strong positive reactions.Western blotting of TLR4showed that the expression of TLR4in peach kernel intervention group(0.5513±0.0432)was higher than that in control group(0.4021±0.0449,P<0.05).Compared with the model group(1.8342±0.0342),the expression of TLR4in peach kernel intervention group decreased significantly(P<0.01).Compared with the control group,the expression of TLR4was significantly increased in the model group(P<0.01).The expression of TRIB3in peach kernel intervention group(1.3638±0.0948)was lower than that in model group(2.3241±0.0654,P<0.01),but higher than that in control group(1.0000±0.0000,P<0.05).The expression of TRIB3in model group was significantly higher than th
