摘 要:目的探究高剂量活血化瘀药丹参、川芎对Lewis肺癌小鼠肿瘤侵袭转移的影响及其可能作用的机制。方法采用腋下接种Lewis肺癌细胞悬液的方法制备Lewis肺癌小鼠模型30只,随机分为模型组、顺铂组、高剂量丹参组、高剂量川芎组、丹参联合顺铂组和川芎联合顺铂组,每组5只。顺铂组、丹参联合顺铂组和川芎联合顺铂组均给予顺铂4 mg/kg腹腔注射,高剂量丹参组、丹参联合顺铂组以2. 16 g/kg剂量丹参灌胃,高剂量川芎组、川芎联合顺铂组以3. 6 g/kg剂量川芎灌胃,模型组给予等体积生理盐水腹腔注射和灌胃。灌胃1次/d,腹腔注射于造模后第1、3、5天进行。每2 d记录1次小鼠体重,并于治疗后第14天将小鼠断颈处死取材,称取肿瘤质量,计算抑瘤率;采用Western-Blot法检测肿瘤组织内Twist和血管内皮生长因子(VEGF)的表达。结果各组小鼠体重,模型组、高剂量丹参组和高剂量川芎组>顺铂组、丹参联合顺铂组和川芎联合顺铂组(P <0. 05),高剂量丹参组、高剂量川芎组和模型组比较差异无统计学意义。计算抑瘤率,发现中药对肿瘤生长并没有抑制作用,反而具有促进作用。肿瘤组织免疫印迹结果显示高剂量川芎组肿瘤组织Twist蛋白表达高于模型组(P <0. 05),高剂量丹参组肿瘤组织VEGF-A蛋白表达高于模型组(P <0. 05)。结论高剂量活血化瘀中药川芎、丹参对Lewis肺癌小鼠肿瘤侵袭转移具有促进作用,其机制可能与调控Twist和VEGF-A蛋白的表达有关。Objective To explore the effects of danshen and chuanxiong,at high dose,in the category of activating blood circulation and resolving stasis and the potential mechanism on tumor invasion and metastasis in Lewis lung cancer mouse.Methods Using the subaxillary inoculation with Lewis lung cancer cell suspension,30mouse models of Lewis lung cancer rats were prepared.They were randomized into a model group,a cisplatin group,a high-dose danshen group,a high-dose chuanxiong group,a danshen plus cisplatin group and a chuanxiong plus cisplatin group,5mouse in each one.In the cisplatin group,the danshen plus cisplatin group and the chuanxiong plus cisplatin group,the intraperito-neal injection with cisplatin,4mg/kg was given.In the high-dose danshen group and the danshen plus cisplatin group,the gavage with danshen,2.16g/kg was given.In the high-dose chuanxiong group and the chuanxiong plus cisplatin group,the gavage with chuanxiong,3.6g/kg was given.In the model group,the normal saline of the same volume was used for in-traperitoneal injection and gavage.The gavage was given once a day and the intraperitoneal injection was on day1,3and5after modeling success.Once every2days,the body weight was recorded.On day14after treatment,the mouse were sacrificed by neck breaking for sample collection.The tumor mass was weighted and the tumor inhibition rate was calculated.The Western-blot method was adopted to determine Twist and VEGF expressions in tumor tissue.Results The body weights in the model group,the high-dose danshen group and the high-dose chuanxiong group were higher than the cisplatin group,the danshen plus cisplatin group and the chuanxiong plus cisplatin group(P<0.05).The difference was not significant between the Chinese herbal medicine group and the model group.It was discovered that Chinese herbal medicine has no inhibitive effect on tumor growth,and instead,it presents the promoting action.The tumor immunity blotting results showed that the Twist protein expression in the high-dose chuanxiong group was higher than
