摘  要:通过比较培养基组成、培养温度、培养基初始pH、光照对茯苓(Wolfiporia cocos)产孢的影响,探讨产孢量较大的培养基组成和培养条件。结果表明:装有60 mL茯苓PDA培养基(200 g马铃薯,20 g葡萄糖,1 g KH_2PO_4,0.5 g MgSO_4·7H_2O,10 mg维生素B_1,18 g琼脂,pH自然,加蒸馏水补足至1000 mL)的350 mL组培瓶,培养基初始pH为6.5,28℃暗培养7 d,12 h光暗交替培养为最优产孢条件,在此条件下,茯苓培养38 d产孢量为(8.12±0.11)×10~9个/瓶。In order to explore optimal medium and culture conditions for sporulation of Wolfiporia cocos,effect of medium component, temperature, initial pH after autoclave and light condition were studied. The results showed that the optimal conditions for sporulation were PDA medium(350 mL with 60 mL medium)composed of 200 g/L potato, 20 g/L glucose, 1 g/LKH2 PO4, 0.5 g/L MgSO4·7H2O, 10 mg/L VB1, and18 g/L agar with natural initial pH at 6.5, cultivation at 28 ℃ in the dark for 7 days followed with a light cycle of 12 h/12 h(irradiation/in the dark). Spore yield under the optimal condition was(8.12 ± 0.11) x 109 per flask on day 38.
