杂交鲟鱼中草药免疫增强剂的体外快速筛选研究

摘　　要：试验抽取杂交鲟鱼(西伯利亚鲟♂×史氏鲟♀)的外周血样,提取外周血白细胞,用浓度0.5 g/m L的苦参、茵陈、杜仲、霪羊藿、茯苓、紫草、甘草、生地8种中草药水提液与外周血白细胞共同孵育后,分别采用抗超氧阴离子试剂盒、金黄色葡萄球菌法测定外周血白细胞的氧呼吸爆发活性及吞噬活性。结果:各处理组外周血白细胞氧呼吸爆发活性均存在显著差异(P<0.05),其中苦参、茵陈、杜仲、霪羊藿、茯苓、紫草、甘草组显著高于对照组(P<0.05),生地组显著低于对照组(P<0.05)。茵陈、杜仲组外周血白细胞吞噬百分比显著高于对照组(P<0.05)。苦参、生地组外周血白细胞吞噬指数显著高于对照组(P<0.05)。结论:综合各评价指标,生地更适合作为杂交鲟鱼的候选免疫增强剂。The aim of this study was to examine the effect of the eight Chinese herbal medicine on the immune protection in Hybrid Sturgeon(Acipenser baerii♂×Acipenser schrenckii♀).Chinese herbal Immunopotentiator were screened using colorimetry method by incubating the isolated blood cell with water extracts of Chinese herbs.Phagocytizing Staphylococcus aureus was used to investigate the effect of water extracts on the respiratory burst activity of the while blood cells on the phagocytic activity.Results:that extracts Sophora flavescens Ait,Artemisiae Scopariae Herba,Eucommia ulmoides Oliv,Epimedii Folium,Poria cocos,Arnebia euchroma could significantly increased the respiratory burst of white blood cells(P<0.05),Rehmannia glutinosa Libosch could significantly decreased the phagocytic index(P<0.05).Artemisiae Scopariae Herba,Eucommia ulmoides Oliv could significantly increased the hagocytic activity(P<0.05).Sophora flavescens Ait and Rehmannia glutinosa Libosch could significantly increased the phagocytic index(P<0.05).In conclusion,Rehmannia glutinosa Libosch were suitable for Hybrid Sturgeon as Chinese herbal medicine Immunopotentiator.
