摘  要:为了验证分泌型卷曲相关蛋白4(secreted frizzled-related protein 4,sFRP4)在肉鸡胫骨软骨发育不良(tibial dyschondroplasia,TD)的胫骨生长板中的表达,以及探究相应预测蛋白的性质与作用,试验采用real-time PCR技术对肉鸡TD生长板中sFRP4的mRNA表达进行检测,并通过生物信息学分析其预测蛋白的性质与作用。结果表明,real-time PCR检测结果与基因芯片结果相匹配,在诱导TD后sFRP4的mRNA表达显著上调。sFRP4基因共编码331个氨基酸,理论等电点为9.50;平均疏水性-0.582,1个糖基化位点,13个磷酸化位点;有信号肽,无跨膜区;二级结构以α螺旋(α-helix)、无规则卷曲(random coil)为主;sFRP4可与Frizzled(Fz)蛋白竞争性结合Wnt8A、Wnt8B、Wnt9A、Wnt10A等蛋白。说明sFRP4蛋白可能是通过抑制Wnt信号通路而影响正常软骨内骨化。To verify the expression of secreted frizzled-related protein4(sFRP4)in the growth plate of tibial dyschondroplasia(TD)and to explore the effects of the predicted sFRP4protein,differentially expressed gene sFRP4was identified by real-time PCR and was analyzed by bioinformatics.The results showed that the results of differential expressed sFRP4were consistent with the results of microarray data.Bioinformatics results showed that the sFRP4gene encoded331amino acids in which theoretical pI=9.50,average hydrophobicity value=-0.582,and O-glycosylation sites and phosphorylation sites of the protein were1and13respectively.The protein had no transmembrane domain,but it had signal peptide.Main structures of the sFRP4protein wereα-helix and random coil.Protein interaction showed that sFRP4and Fz protein can competitively combine with Wnt8A,Wnt8B,Wnt9A,Wnt10A etc.
