摘  要:通过腹腔注射四氯化碳(CCl4)建立小鼠急性肝损伤模型,研究黄芪粗提物对CCl4诱导的小鼠急性肝损伤的保护作用。设空白组、模型组、阳性组和黄芪粗提物高、中、低剂量组(8、4、2g/kg),连续灌胃给药15d,末次给药后腹腔注射CCl4,分别测定小鼠肝脏指数,血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)、丙二醛(MDA)及肝脏中甘油三酯(TG)的含量,观察小鼠肝脏病理组织学变化。与模型组比较,黄芪粗提物高、中、低剂量组显著或极显著降低肝脏指数(P<0.01或P<0.05),降低血清中ALT、AST、MDA及肝脏中TG的含量(P<0.01或P<0.05),显著或极显著减轻小鼠肝纤维化的程度(P<0.05,P<0.01)。结果表明,黄芪粗提物对CCl4诱导的小鼠急性肝损伤有保护作用,可为黄芪粗提物在临床上的应用提供理论基础。The model of acute liver injury was established by intraperitoneally injecting CCl4in mice,the protective effect of Astragalus crude extracts on CCl4-induced acute liver injury in mice was investigated.The mice were divided into blank group,model group,positive group and Astragalus crude extracts high,medium and low dose groups(8,4,2g/kg),drugs were administered continuously for15d,the CCl4was intraperitoneally injected after the last administration.The liver index,ALT,AST and MDA in sera,TG in liver tissue were measured.The hepatic histopathological lesions were observed in mice.The results showed that compared with model group,the Astragalus crude extracts high,medium and low dose groups could significantly decrease the liver index of mice(P<0.01or P<0.05),ALT,AST and MDA contents in sera and TG content in liver tissue(P<0.01or P<0.05)and significantly reduced the degree of liver fibrillation in mice.The Astragalus extracts have protective effect on CCl4-induced acute liver injury in mice,which provided a theoretical basis for the clinical application.
