摘　　要：为探讨FAR1在不同等级卵泡和不同就巢时期卵泡中的表达规律,实验通过RT-PCR、RACE技术克隆FAR1基因mRNA序列,并用RT-qPCR检测浙东白鹅不同等级卵泡以及不同就巢时期卵泡的FAR1表达。结果表明:FAR1 mRNA包含83 bp的5'UTR,1 557 bp的开放阅读框(ORF)和229 bp的3'UTR,共编码518个氨基酸;系统进化树分析显示,浙东白鹅与鸭、鸡三者聚为一类;RT-qPCR检测显示,等级卵泡中FAR1表达量显著高于等级前卵泡(P<0.05),但F4、F5等级卵泡低于卵巢组织表达量,产蛋期FAR1表达量显著高于就巢期(P<0.05)。因此推测,FAR1基因与鹅生殖调控密切相关,不仅参与了鹅卵泡发育,且在产蛋期高表达。To investigate the expression of FAR1 in follicles on different levels and different incubation period.In this study,the FAR1 mRNA sequence was cloned by reverse transcription polymerase chain reaction(RT-PCR)and rapid-amplification of cDNA ends(RACE),and detected the expression of FAR1 in follicles on different levels and different incubation period of the Zhedong goose by RT-qPCR.The results showed that the FAR1 cDNA contains a 83bp 5'UTR,a 1 557 bp open reading frame(ORF)and a 229 bp 3'UTR,encodes 518 amino acids;The phylogenetic tree analysis showed that the eastern Zhedong goose,duck and chicken were clustered into one category.RT-qPCR detection the results showed that the expression level of FAR1 in graded follicles was significantly higher than that in pre-grade follicles(P<0.05),but that in F4 and F5 grade follicles were lower than that in ovarian tissue.At the same time,it was also found that the expression of FAR1 in the laying period was significantly higher than that in the incubation stage(P<0.05).Therefore,FAR1 gene is closely related to the regulation of goose reproduction.It not only participates in the follicle development of the geese,but also is highly expressed during the laying period.
