摘  要:目的探讨不同性别大鼠在模拟海拔8000 m环境下不同时间生理生化指标的变化及差异。方法将100只大鼠(雌雄各半)随机分为正常组、缺氧1、3、5和10 d组,采用大型低压氧舱模拟海拔8000 m高原环境,测定大鼠血液流变学指标和脑组织生化指标,并对脑和心肌进行组织病理观察。结果各缺氧组大鼠脑和心肌组织均有损伤,且以缺氧10 d组的损伤最为严重。与正常组比较,各缺氧组大鼠全血黏度、卡松黏度、屈服应力以及脑组织丙二醛均显著升高(P <0. 05,P <0. 01),且随缺血时间延长逐渐升高;各缺氧组大鼠脑组织超氧化物歧化酶、过氧化氢酶、谷胱甘肽过氧化物酶、一氧化氮和一氧化氮合成酶显著降低(P <0. 05,P <0. 01),且随缺氧时间延长逐渐降低。雌鼠和雄鼠在上述指标比较时具有一定差异。结论高原缺氧可对大鼠的脑和心组织损伤,生理生化指标明显改变,将严重影响大鼠正常的生理代谢。Objective To investigate changes and differences of physiological and biochemical indexes at different times in simulated altitude 8000 m environment in different gender rats.Methods A total of 100 rats(50 males and 50 females)were randomly divided into normal group,hypoxia for 1,3,5 and 10 d groups.Large hypobaric oxygen cabin was used to simulate high altitude environment at 8000 m.Hemorheological indexes and biochemical indexes of brain tissue were detected,and histopathological changes of brain and myocardium tissues were observed in rats.Results Brain and myocardium tissues were all damaged in all hypoxia groups,and the injury was most serious in hypoxia for 10 d group.Compared with those in normal group,levels of whole blood viscosity,Casson viscosity,yield stress and malondialdehyde in brain tissue of rats in each hypoxic group were significantly increased(P<0.05,P<0.01),and the levels were gradually increased with prolonging ischemic time;levels of superoxide dismutase,catalase,glutathione peroxidase,nitric oxide and nitric oxide synthetase of rats'brain tissues in each hypoxic group were significantly decreased(P<0.05,P<0.01),and the levels were gradually decreased with prolonging hypoxia time.There were differences in the above indexes between female with male rats.Conclusion Highland hypoxia may damage brain and cardiac tissues of rats,and change the physiological and biochemical indexes are significant,which will seriously affect normal metabolism of rats.
