摘  要:目的探讨特发性肺纤维化(IPF)大鼠机体氧化损伤的特点和益气养阴祛瘀方药的作用机理。方法135只Wistar大鼠分为对照组、模型组、低剂量组、高剂量组和强的松组,每组27只。大鼠IPF造模后第2天开始给药,每日1次,并于给药7、14和28 d观察各组大鼠血清和肺组织的丙二醛(MDA)含量与超氧化物歧化酶(SOD)活力变化以及肺组织病理学形态变化。结果模型组的血清和肺组织MDA含量普遍高于对照组(P <0. 05),SOD活性低于对照组(P <0. 05);高、低剂量组和强的松组的血清和肺组织MDA含量普遍低于模型组,SOD活性普遍强于模型组(P <0. 05);高、低剂量组的肺组织SOD活性强于强的松组(P <0. 05)。高、低剂量组及强的松组肺泡间隔增宽,炎性细胞浸润程度低于模型组,成纤维细胞少量增殖,其中,高剂量组大鼠炎性细胞浸润及肺纤维化程度最轻。结论益气养阴祛瘀方药可清除IPF大鼠体内MDA,增强SOD活性,有效减轻组织氧化损伤及肺纤维化程度。Objective To investigate characteristics of oxidative damage in rats with idiopathic pulmonary fibrosis(IPF)and action mechanism of Supplementing Qi and Nourishing Yin Prescription.Methods A total of 135 Wistar rats were divided into control group,model group,low dose group,high dose group and Prednisone group(n=27 for each group).The rats were administered once a day on the 2 nd d after having IPF modeling,and contents of malondialdehyde(MDA)in serum and lung tissues,activity of superoxide dismutase(SOD)and histopathologic changes of lung tissues were observed on the 7 th 14 th and 28 th d after administration.Results In model group,MDA contents in serum and lung tissue were significantly higher(P<0.05),while SOD activity was significantly lower than those in control group(P<0.05).In high,low dose and Prednisone groups,MDA contents in serum and lung tissue were significantly lower,while SOD activities were stronger than those in model group(P<0.05).SOD activities in lung tissue in high and low dose groups were stronger than that in model group(P<0.05).In high,low dose and Prednisone groups,values of alveolar septum were significantly increased,while degrees of inflammatory cell infiltration were significantly lower than those in model group(P<0.05).Fibroblasts were proliferated slightly,and degrees of inflammatory cell infiltration and pulmonary fibrosis were the least in high dose group.Conclusion Supplementing Qi and Nourishing Yin Prescription may eliminate MDA in vivo in IPF rats,enhance SOD activity and effectively decrease degrees of oxidative damage and pulmonary fibrosis.
