摘  要:为提高废次烟叶中绿原酸的提取率,本研究以废次烟叶为材料,优化绿原酸的提取工艺。采用单因素试验,研究液料比、浸提温度、浸提时间、溶剂pH对绿原酸提取率的影响。在此基础上,用L9(34)正交试验优选废次烟叶绿原酸提取工艺条件。采用最佳优选工艺组合提取不同品种烟草不同部位的绿原酸,并研究提取液的抗氧化性。结果表明:随着液料比的增大、浸提温度的升高、浸提时间的延长、溶剂pH的增大,废次烟叶中绿原酸提取率均呈先上升后下降趋势。通过方差分析和多重对比发现,这4个因素对绿原酸提取率均有极显著影响,最佳优选工艺组合是浸提时间为90min、浸提温度为90℃、液料比为10∶1、pH5时,提取率为0.714%。通过验证试验表明,这种优选工艺组合提取率高,稳定性好。运用此方法,对不同品种烟草不同部位的绿原酸进行提取,发现不同部位间提取率差异显著,上部叶最高,其次是中部叶和下部叶,烟梗的提取率最低,且品种间提取率也存在显著差异。此外,废次烟叶提取液具有良好的清除DPPH自由基能力和Fe3+还原能力,具有较高的潜在应用价值。In order to improve the extraction rate of chlorogenic acid in waste tobacco leaves,the discarded tobacco leaves were used to establish the best technology combination for extracting chlorogenic acid in this study.Using single-factor experiments,the effects of liquid-feed ratio,extraction temperature,extraction time,and solvent pH on the extraction of chlorogenic acid were studied.On this basis,the extraction conditions of chlorogenic acid from the discarded tobacco leaves were optimized by L9(34)orthogonal experiment.Extracting chlorogenic acid from different parts and different parts using the best combination of process techniques,and studying the antioxidant activity of the extract.The results showed that with the ratio of liquid to material,extraction temperature,and extraction time as well as solvent pH increasing,the extraction rate of chlorogenic acid in the discarded tobacco leaves increased firstly and then decreased.Through analysis of variance and multiple comparisons,it was found that the four factors had a significant effect on the extraction rate of chlorogenic acid.The best optimized extraction time was90min,extraction temperature90℃,liquid-to-feed ratio10∶1,solvent pH5.Under this condition,the extraction rate was0.714%.The verification experiments showed that this optimal combination had a high extraction rate and a strong stability.Using this method,the extraction of chlorogenic acid from different parts of tobacco was studied.It was found that the highest extraction rate was found in the upper leaves,followed by the middle and the lower leaves,and the extraction rate of stems was the lowest.In addition,the waste secondary tobacco leaf extract has a good DPPH free radical scavenging and reducing ability and has a high potential application value.
