摘  要:从河北省发病鸵鸟采集病料,经鸡胚传代、血清学试验、毒力指标测定,分离鉴定了1株新城疫病毒(Newcastle disease virus,NDV)(命名为HB/1/08/Os株),对其进行了核衣壳蛋白基因遗传进化分析和人工感染试验。结果表明,分离毒株的致死鸡胚平均死亡时间为55.2h,1日龄雏鸡脑内接种后致病指数为1.88。该病毒NP基因含有1个长为1470bp的开放阅读框,编码489个氨基酸。同源性分析结果表明,该毒株的NP基因与35株参考毒株间的核苷酸和氨基酸的同源性分别为77.3%~98.4%和90.6%~99.6%。遗传进化分析表明,该毒株与基因Ⅶd亚型毒株ZJ1关系最近,与疫苗株和I类毒株关系较远。综合判定该毒株为强毒。因此,须加强河北省鸵鸟NDV的监测。A Newcastle disease virus(NDV)(named HB/1/08/Os)was identified by chicken embryo passage,serological test,and pathogenicity index tests from diseased ostrich sample in Hebei province.Genetic analysis of nucleocapsid protein(NP)gene and artificial infection study were carried out.Pathogenicity indexes such as the mean death time(MDT)and intracerebral pathogenicity index(ICPI)were 55.2 h and 1.88,respectively.Sequence analysis indicated the NP gene contains 1 open reading frames composed of 1470 nucleotides,and encoded 489 amino acids.The homology analysis showed that the nucleotide and amino acid homology was 77.3%98.4%and 90.6%99.6%between the NP gene of the virus and the 35 reference strains,respectively.Phylogenetic analysis based on the NP gene nucleotide sequence revealed that this isolate belonged to genotypeⅦd,had the closest relationship with the strain ZJ1,long relationships with the vaccine strains and the class I strains.The results revealed the isolate to be a velogenic NDV.Therefore,it is necessary to strengthen the monitoring of ostrich NDV in Hebei province.
