摘 要:采用拉曼光谱法对药用植物丹参根鲜样不同组织部位进行原位检测,以考察丹参酮类成分在丹参根中不同组织上的分布。结果表明:丹参酮IIA标准品在1668、1620、1579、1535、1479cm-1出现较强峰,丹参根周皮、韧皮部也在1667/1665、1618/1613、1576/1575、1533/1532、1477/1480cm-1出现较强峰,周皮、韧皮部与丹参酮IIA标准品谱图之间的匹配度分别为74.9%、49.4%;而木质部与丹参酮IIA标准品无相对应的拉曼峰,与隐丹参酮、二氢丹参酮标准品谱图之间的匹配度很低,分别仅为4.4%、5.6%,不足以证明有丹参酮类成分存在;因此得出丹参根周皮和韧皮部中均有丹参酮类成分的分布,且主要成分为丹参酮IIA,而木质部中无丹参酮类成分分布的结论。虽然丹参根周皮和木质部均呈红色,但拉曼光谱检测发现丹参酮类成分的分布与周皮细胞颜色分布之间呈正相关性,与木质部细胞颜色分布无相关性,这提示颜色判别法分析丹参酮类成分的分布具有一定的局限性,而利用拉曼光谱法可快速的获得丹参酮类成分在丹参根中的分布情况,凸显了拉曼光谱用于丹参根中化学成分原位检测的优势。Raman spectroscopy was used to investigate the tissue-specific distribution of tanshinones in different tissue of Danshen in situ.The results showed that there were strong peaks of tanshinone IIA at1668,1620,1579,1535,1479cm^-1.And in the periderm,phloem of Danshen’s roots at1667/1665,1618/1613,1576/1575,1533/1532,1477/1480cm^-1respectively.The matching degree between Raman spectrum of periderm,phloem of Danshen’s roots and tanshinone IIA referenced spectrum were74.9%and49.4%respectively.While there was no relationship between the Raman spectrum of xylem and tanshinone IIA referenced spectrum,and the matching degree between Raman spectrum of xylem of Danshen’s roots and cryptotanshinone,dihydrotanshinone referenced spectrum was only4.4%and5.6%respectively.Therefore,tanshinones was mainly distributed in periderm,phloem of Danshen’s roots,and the main component was tanshinone IIA,but no distribution of tanshinones was found in xylem.Although the color of periderm and xylem of Danshen’s root both looked like red,our study showed that there was a positive correlation between the distribution of the tanshinones and the color distribution of the periderm cells but no correlation with the color distribution of the xylem cells.This suggests that the mothed of color discrimination to analysis of the distribution of tanshinones has certain limitations.The distribution of tanshinones in Danshen’s roots can be quickly obtained by Raman spectroscopy,highlighting the advantages of Raman spectroscopy in detection of chemical constituents of Danshen’s roots in situ.
