摘 要:应用电化学法研究低浓度葡萄糖对PC12细胞的影响。研究PC 12细胞的电化学行为,并通过标准品对比、高效液相法对细胞的电化学信号进行归属。将PC12细胞分为4. 5mg/ml葡萄糖阳性对照组、无糖阴性对照组和0. 2mg/ml低糖组,采用电化学法研究低浓度葡萄糖对PC12细胞的嘌呤核苷酸代谢的影响。得出PC12细胞的三个电化学信号分别来源于鸟嘌呤和黄嘌呤的混合以及次黄嘌呤和腺嘌呤的混合及其终产物尿酸。低浓度葡萄糖对PC12细胞活性有损害作用,且影响其嘌呤核苷酸代谢,存在糖浓度-效应关系和时间-效应关系。Objective:To study the effect of low concentration glucose on PC12 cells.METHODS:The electrochemical behavior of PC12 cells was studied,and the electrochemical signals of cells were assigned by standard comparison and high performance liquid chromatography.PC12 cells were divided into 4.5mg/ml glucose positive control group,glucose-free control group and 0.2mg/ml low glucose group.The effect of low concentration glucose on the purine nucleotide metabolism of PC12 cells was studied by electrochemistry.RESULTS:The three electrochemical signals of PC12 cells were derived from the mixture of guanine and xanthine,the mixture of hypoxanthine and adenine and the final product uric acid.Conclusion:Low concentration of glucose has a detrimental effect on the activity of PC12 cells,and affects its purine nucleotide metabolism.There are sugar concentration-effect relationship and time-effect relationship.
