摘　　要：CpG-DNA是一些具有免疫激活功能的以未甲基化的CpG基序为核心的DNA序列,能刺激多种免疫活性细胞,诱导机体天然免疫系统活化。本试验依据CpG-DNA的特性,经过合成和克隆的方法制备出32条CpG-DNA序列,通过淋巴细胞增殖试验筛选出4种对羊、小鼠淋巴细胞都有较强细胞增殖作用的序列,将淋巴细胞增殖试验选出的CpG进行动物安全性试验,结果均无毒副作用,为进一步开展CpG免疫增强剂免疫效力试验奠定了基础。CpG-DNAis the core of non-methylated DNAsequence.It has immune activation function,which can stimulate variety of immune cells,inducing the activation of the body's natural immune system.According to the characteristics of CpG-DNA,32CpG-DNA sequences were cloned and synthesized.These sequences were screened by the lymphocyte proliferation test,and find4kinds of sequences which have strong cell proliferation effect on the sheep and mice lymphocytes.The4kinds of sequences were used to carried out the animal safety test.The result showed that the4kinds of sequences do not have toxic and side effects.which laid a foundation for the immune efficacy test of CpG-DNA immune enhancer.
