牛蒡子总木脂素和淫羊藿总黄酮配伍对2型糖尿病大鼠骨质疏松的保护作用

摘　　要：目的探讨牛蒡子总木脂素、淫羊藿总黄酮及其两种不同比例配伍对2型糖尿病大鼠骨质疏松的保护作用。方法采用高脂饲料喂养联合小剂量链脲佐菌素(STZ)诱导2型糖尿病大鼠骨质疏松模型,实验分为6组,正常组、模型组、牛蒡子总木脂素组、淫羊藿总黄酮组、牛蒡子总木脂素-淫羊藿总黄酮1.5∶1.0配伍组和1.0∶1.5配伍组(质量比)。正常组和模型组给予生理盐水,其他组分别给予相应剂量的药物,每天灌胃给药1次,连续12周后,麻醉,腹主动脉取血,测血清胰岛素(INS)、总胆固醇(TC)、三酰甘油(TG)、血钙(Ga)、血磷(P)、骨钙素(BGP)等指标;并取双侧股骨,测定骨干及骨灰重,骨钙及骨磷的含量,以及骨最大载荷、最大能量吸收值、骨结构硬度等骨生物力学指标。结果各给药组均能不同程度地改善2型糖尿病骨质疏松大鼠的血清胰岛素等糖脂代谢指标,提高血钙、血磷和骨钙素水平,增加骨干重、骨灰、骨钙、骨磷等骨矿物质含量;淫羊藿总黄酮组及两配伍组可改善骨生物力学指标(P<0.05或P<0.01)。其中,与单药组相比,配伍组对提高胰岛素、骨钙素、骨钙、骨磷水平,改善骨硬度作用更佳,牛蒡子总木脂素-淫羊藿总黄酮1.0∶1.5配伍组作用尤为显著(P<0.05或P<0.01)。结论牛蒡子总木脂素与淫羊藿总黄酮配伍可明显改善2型糖尿病大鼠骨质疏松,开发牛蒡子总木脂素、淫羊藿总黄酮配伍的中药复方新药,可为糖尿病骨质疏松并发症的防治提供新用药。Objective To explore the protective effects of Arctiin total lignans,Epmedium Flavonoids and their two different compatibilities on osteoporosis in SD rats with type2diabetes mellitus.Methods Rats were fed with high-fat diets and treated with35mg/kg streptozotocin-induced osteoporosis caused by type2diabetics.The rats were divided six groups,including the normal control group,model group,Arctiin total lignans group,Epmedium Flavonoid group,Arctiin total lignans-Epmedium Flavonoi1.5∶1.0compatibility group and1.0∶1.5compatibility group.The normal control and the model group were given saline by intragastric administration,and the other groups were given the appropriate dose of the corresponding drugs once a day.After12weeks,blood was collected from the abdominal aorta under anesthesia,and the serum levels of insulin(INS),total cholesterol(TC),triglyceride(TG),calcium(Ca),phosphorus(P)and bone gla protein(BGP)were determined.In addition,bilateral femurs were collected to measure backbone and bone ash,bone calcium and bone phosphorus content,as well as bone biomechanics.Results All treatment groups showed an improvement of the serum levels of INS,TC,TG,Ca,P and BGP,and an increase of backbone and bone ash weight,bone calcium and bone phosphorus content,as compared to the model group(P<0.05or P<0.01).The Epmedium Flavonoids and the two compatibility groups enhanced the biomechanical index of bone.The1.0∶1.5compatibility group,especially,demonstrated better effect than both Arctiin total lignans and Epmedium Flavonoids(P<0.05or P<0.01).Conclusion The combination of Arctiin total lignans and Epmedium Flavonoids shows a significant effect on improvement of osteoporosis in type2diabetes mellitus,which may be develop a new compound Chinese medicine for the prevention and treatment of complications of diabetic osteoporosis.
