摘  要:目的研究探讨艾瑞昔布不同用药持续时间对大鼠股骨骨折愈合的影响及与BMP-2、VEGF表达的关系。方法 75只SD大鼠构建闭合性股骨骨折,实验随机分为正常组(生理盐水灌胃4周);短期用药组(艾瑞昔布混悬液2 mg/(kg·d),灌胃2周后改用生理盐水灌胃2周);长期用药组(艾瑞昔布混悬液2 mg/(kg·d),灌胃4周),放射性(X线)评估、骨痂骨密度(BMD)测量、血清BMP-2、VEGF浓度测定、HE染色和免疫组化染色评估。结果短期用药组和长期用药组骨折愈合评分、骨痂BMD、血清BMP-2和VEGF浓度、BMP-2和VEGF阳性表达均降低,其中尤以长期用药组降低最显著(P <0.05)。短期用药组骨小梁生长稍稀疏,可见软骨细胞,成骨细胞相对较少,骨痂见大量软骨组织;长期用药组骨小梁生长稀疏,可见大量软骨细胞,成骨细胞生长少见,骨痂见大量纤维组织。结论艾瑞昔布对大鼠骨折愈合有抑制作用,其中以长期用药抑制作用最明显,其可能与降低BMP-2、VEGF表达有关。Objective To investigates the effect of different durations of Imrecoxib on fracture healing and its relationship with the expression of BMP-2 and VEGF in rats.Methods Closed femoral fractures were estab-lished in 75 rats,andthey were randomly divided into normal group(saline continued gavage 4 weeks);short-term medication group(Imrecoxib suspension 2 mg/(kg·d),continued gavage 2 weeks,switch to saline 2 weeks)and-long-term medication group(Imrecoxib suspension 2 mg/(kg·d),continuous gavage 4 weeks).Radioactive(X-ray)assessment,callus bone mineral density(BMD)measurement,serum BMP-2 and VEGF concentration were measured and determined and HE staining and immunohistochemical staining were conducted.Results Fracture healing score,osteophyte BMD,serum BMP-2 and VEGF concentrations,BMP-2 and VEGF positive expression were decreased in the short-term and long-term medication group and the most significant reduction was observed in the long-term medication group(P<0.05).Trabecular bone growth was slightly sparse in short-term medication group,showing chondrocytes,relatively less osteoblasts,and a lot of callus cartilage tissues.Inlong-term medica-tion group,trabecular bone growth was slightly sparse,showing a large number of chondrocytes,rare osteoblastic growth,and a large number of fibrous callus tissues.Conclusion The healing of fracture in rats is significantly inhibited by Imrecoxib,and the effect is most ficant with a long-term medication,which may be related to the reduction of BMP-2 and VEGF expression.
