摘  要:为了给生物质水解液研究中的甲酸含量检测提供可靠的方法,建立了反相高效液相色谱法(RP-HPLC)分析生物质水解液中甲酸含量的方法。运用Ultimate LP-C18色谱柱(250mm×4.6mm,5μm),以V(NaH_2PO_4水溶液)∶V(甲醇)=97∶3[c(NaH_2PO_4)=0.02mol/L(pH=2.5)]体系为流动相,在流速1.0mL/min、柱温30℃、检测波长210nm条件下,实现了生物质水解液中甲酸的分离分析。方法评价结果表明该法测定甲酸的线性范围为0.01~0.10mol/L(相关系数R^2=0.998 7),甲酸平均回收率为96.6%~105.0%,RSD值为1.4%~2.5%(n=3)。该法测定生物质水解液中的甲酸具有操作简单易行、分析结果准确可靠的优点,可作为生物质水解液中甲酸含量分析的常规方法。A method was developed for the determination of formic acid in biomass hydrolysate by Reversed-phase high performance liquid chromatography(RP-HPLC).Ultimate LP-C18(250 mm×4.6 mm,5μm)was used as the analytical column.The mobile phase was methanol-0.02 mol/L sodium dihydrogen phosphate(V/V=97∶3),pH=2.5 and the column temperature is 30 degrees.The flow rate was 1.0 mL/min,and detection wavelength was 210 nm.The results show that the linear range of the method was 0.01~0.10 mol/L with good linear relationship(R 2=0.998 7).The average recovery was 96.6%~105.0%.The date for formic acid determined is repeatable with RSD of 1.4%~2.5%(n=3).The method is simple,realizable,accurate and reliable and can be used as a conventional method for determination of formic acid in biomass hydrolysate.
