摘 要:目的观察活血解毒中药组分配伍对急性心肌梗死(acute myocardium infarction,AMI)后大鼠不同时段炎症水平的干预作用。方法体重180~200 g的Wistar大鼠60只,结扎前降支中上1/3处造成AMI模型;假手术组仅穿线不结扎。造模24小时后存活的大鼠随机分为6组,每组5只,即空白对照(假手术)组、AMI(模型)组、阳性药物对照组(辛伐他汀)组、活血药(赤芍总苷)组、解毒药(黄连生物碱)组、活血解毒药(赤芍总苷+黄连生物碱)组。各组给予药物或者等量生理盐水灌胃治疗。各组分4个时间段:均连续灌胃1天、3天、7天和14天,动态测定大鼠血清白细胞介素-1β(interleukin-1β,IL-1β),肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、高敏C-反应蛋白(high-sensitivity,hs-CRP)和单核细胞趋化蛋白-1(monocyte chemotactic protein 1,MCP-1)的变化。结果(1)造模后第1天,模型组大鼠血清hs-CRP、IL-1β、TNF-α和MCP-1水平开始升高,和假手术组比有显著差异(P <0. 05);和模型组比,活血组大鼠血清MCP-1水平、活血解毒组大鼠TNF-α有明显抑制作用(P <0. 05);(2)造模后第3天,模型组大鼠血清炎症因子含量显著升高,和假手术组比较有显著差异(P <0. 05);与模型组相比,活血组、解毒组、活血解毒组分配伍对大鼠血清hsCRP、IL-1β、hs-CRP和MCP-1含量有不同程度的抑制作用(P <0. 05);(3)造模后第7天,大鼠血清炎症因子含量较第3天有所下降,但仍明显高于假手术组(P <0. 05);和模型组相比,辛伐他汀组、活血组及活血解毒组对TNF-α均有明显抑制作用;解毒组和活血解毒组对大鼠血清IL-1β含量有显著抑制作用(P <0. 05);(4)造模后第14天,模型组大鼠血清MCP-1和IL-1β水平再次上升,与AMI后第3天水平相当; hs-CRP和TNF-α含量较前略有下降,和假手术组比较均有显著差异(P <0.05)。和模型组相比,各用药组均不同程度地呈现出对大鼠血清hs-CRP、IL-1β、TNF-α和MCP-1的抑制作用(P Objective To observe the effects of component compatibility of promoting blood circulation and detoxification herbs on inflammation level after acute myocardial infarction(AMI)in different periods.Methods Wistar rats weighing180~200g were subjected to ligate the middle and upper1/3of the anterior descending artery to create an AMI model,while in the sham group,only threading was performed,not ligation.All the survival AMI rats after24hours were divided into6groups with5rats in each group.The6groups were sham operation group,model group,simvastatin group,Huoxue herb group,Jiedu herb group,Huoxue Jiedu herb group.Each group was given medicine or equal amount of normal saline for gavage.The serum level of interleukin-1beta(IL-1β),tumor necrosis factor alpha(TNF-α),high-sensitivity(hs-CRP)and monocyte chemoattractant protein-1(MCP-1)in rats after continuous intragastric administration for1,3,7and14days.Results(1)1day after modeling,the serum level of IL-1β,TNF-α,hs-CRP and MCP-1increased greatly,which was significantly different from that in the sham group(P<0.05);Compared with the model group,serum MCP-1levels in the Huoxue group and TNF-αin the Huoxue Jiedu group were significantly inhibited(P<0.05);(2)3days after modeling,the content of serum inflammatory factors in model group was significantly increased compared with the sham group(P<0.05);Compared with the model group,the content of IL-1β,TNF-α,hs-CRP and MCP-1decreased in different degrees in Huoxue group,Jiedu group and Huoxue Jiedu group(P<0.05);(3)7days after modeling,serum inflammatory cytokines content decreased comparing with the3days,but still significantly higher than the sham operation group(P<0.05);Compared with the model group,simvastatin,Huoxue herbs and Huoxue Jiedu herbs could inhibit TNF-αobviously while Huoxue Jiedu herbs could inhibit serum IL-1βsignificantly(P<0.05);(4)14days after modeling,the serum MCP-1and IL-1βlevels of model group increased again,which was similar to those of3days after AMI,while hs-CRP and TNF-αdecreased slig
