摘  要:目的:探讨ω-3鱼油脂肪乳对急性一氧化碳(CO)中毒后大鼠血液流变学及红细胞膜流动性的影响。方法:采用腹腔注射法制备急性CO中毒大鼠模型,建模成功大鼠随机分为ω-3鱼油脂肪乳组和生理盐水组,对照组大鼠腹腔注射相同剂量的空气。尾静脉采血,用双球直管式技术测定全血黏度、血浆黏度、血细胞比容、红细胞变形指数及红细胞刚性指数,荧光偏振法测定大鼠红细胞膜偏振度。结果:与生理盐水组比较,鱼油脂肪乳组大鼠1、7、14d大鼠低切全血黏度明显降低(P<0.01)。鱼油脂肪乳大鼠红细胞比容(HCT)较对照组相比未见明显变化,7、14天时较生理盐水组大鼠低,差异有统计学意义(P<0.05)。鱼油脂肪乳大鼠红细胞变形指数(DI)及红细胞刚性指数(IR)较生理盐水组大鼠1、7、14天时变化明显,差异有统计学意义(P<0.05)。脂肪乳组大鼠红细胞膜偏振度(P)较生理盐水组大鼠显著减小(P<0.05)。结论:鱼油脂肪乳可提高一氧化碳中毒大鼠红细胞变形性和降低血液黏度,改善血液流变性,提高红细胞膜流动性,改善微循环减轻组织缺氧状态。Objective:To investigate the effect of ω-3 fish oil fatty emulsion on hemorheology and erythrocyte membrane fluidity in rats after acute carbon monoxide(CO)poisoning.Methods:The rat model of acute CO poisoning was prepared by intraperitoneal injection,which was randomly divided into fish oil fat emulsion injection group and normal saline group,the same dose of air was injected into the rat model of the air control group,and the whole blood viscosity,plasma viscosity,hematocrit,erythrocyte deformation index and erythrocyte rigidity index were determined by double-ball direct tube technique.The polarization degree of erythrocyte membrane in rats was determined by fluorescence polarization method.Results:Compared with the normal saline group,low shear whole blood viscosity of 1,7 and 14d rats in fish oil fatty emulsion group was significantly reduced(P<0.01).There was no significant change in hematocrit(HCT)in fish oil fatty emulsion rats compared with the control group,and7or14days were lower than those in acute CO poisoning group,and the difference was statistically significant(P<0.05).The erythrocyte deformation index(DI)and erythrocyte rigidity index(IR)of fish oil fatty emulsion rats were significantly different in 1,7 and 14 ays in rats with normal saline group,and the difference was statistically significant(P<0.05).The polarization degree(P)of erythrocyte membrane in rats with fat emulsion intervention group decreased significantly(P<0.05)in rats with normal saline group.Conclusion:The fish oil fatty emulsion can increase erythrocyte deformability and reduce blood viscosity,improve blood rheology,fluidity of erythrocyte membrane,microcirculation and reduce the hypoxia state of tissue in rats with carbon monoxide poisoning.
