摘  要:为了研究2709碱性蛋白酶水解猪肠黏膜制备肝素工艺条件,在单因素试验基础上,利用Box-Benken进行试验设计,对水解条件进行了优化研究和验证试验。结果表明,2709碱性蛋白酶水解条件为酶底物质量分数0.70%,水解温度55℃,水解时间3 h,pH值8.8,食盐浓度0.3 mol/L,料液比为1∶7(g∶L);验证试验证明该水解条件是可靠的。In this paper,in order to get the hydrolysis conditions optimization of heparin made from hydrolyzed pig intestine mucosa by2709alkaline protease,the hydrolysis conditions were optimized by the Box-Benken design on the basis of single-factor tests,and the validation experiments were performed.The results showed that the hydrolysis conditions of2709alkaline protease were enzyme substrate ratio0.70%,temperature55℃,hydrolysis time3h,pH8.5,salt concentration0.3mol/L,material liquid ratio1∶7(g∶L),and the validation experiment confirmed that the hydrolysis condition was reliable.
