摘 要:目的探讨大孔吸附树脂纯化分离了哥王中西瑞香素的最佳工艺条件。方法采用高效液相色谱(HPLC)法测定西瑞香素的含量,采用静态吸附和动态吸附结合的方法从8种树脂中优选出一种后对其工艺进行优化。结果 HPD600是纯化了哥王中西瑞香素的优选树脂,最佳吸附条件为上样浓度90μg·m L^(-1),上样液体体积45 m L,上样速度1. 5 m L·min^(-1),吸附率达97. 5%;较佳的洗脱条件:乙醇浓度90%,洗脱剂体积为30 m L,洗脱速度为1.5 m L·min^(-1),解吸率为96.4%。结论大孔吸附树脂HPD600能有效分离纯化了哥王中西瑞香素。Objective To investigate the best purification conditions of the dephnoretin from Wikstroemia indica Linn by macroporous resin.Methods The content of dephnoretin was determined by HPLC method.The8kinds of macroporous resins was selected by static and dynamic adsorption and desorption,and the purification parameters was optimized.Results HPD600was the best resin to purifiy daphne of Wikstroemia indica Linn;The optimum adsorption conditions for the sample concentration was90μg·mL^-1,the upper sample volume was45mL,the sample rate was1.5mL·min^-1,the adsorption rate reached97.5%;the ethanol concentration was90%for better conditions of elution,the eluent volume was30mL,the elution rate was1.5mL·min^-1,the desorption rate was96.4%.Conclusion HPD600macroporous adsorption resin can effectively separate and purify dephnoretin of Wikstroemia indica Linn.
