摘 要:目的:建立用于测定患者血浆中利奈唑胺浓度的高效液相色谱法,为临床个体化给药提供依据。方法 :以奥卡西平为内标物;采用蛋白沉淀法进行前处理,沉淀剂为甲醇;色谱柱为Hypersil ODS C_(18)柱(250 mm×4.6 mm,5μm);流动相:甲醇-水(40:60,v:v);检测波长:254 nm;流速:1.0 mL·min^(-1);柱温:35℃;进样量:20μL。结果:利奈唑胺在0.5~50.0 mg·L^(-1)的范围内线性关系良好(~r2=0.999,n=7);方法回收率和提取回收率分别≥91.69%、≥81.50%;日内、日间精密度的RSD分别≤10.30%、≤11.50%;长期稳定性、反复冻融稳定性、室温稳定性(室温下放置12 h)的RSD分别≤13.52%、≤5.36%、≤10.76%;所测得的患者血浆中利奈唑胺的药物浓度在0.73~6.30 mg·L^(-1)之间。结论:本实验建立的方法操作简单、灵敏快速、准确度高,可用于临床监测患者血浆中利奈唑胺的药物浓度。Objective:To establish the high performance liquid chromatography(HPLC)method for determinating the plasma concentration of linezolid(LZD)in patients,and provide the evidence for individual therapy.Methods:The internal standard substance was oxcarbazepine.Methanol was used as the precipitator for the pretreatment of plasma.The chromatographic column was Hypersil ODS C18(250 mm×4.6 mm,5μm).The mobile phase consisted of methanol and water(40:60).The flow rate was 1.0 mL·min^-1.The column temperature was 35℃.The sample size was 20μL.Results:LZD showed a good linear relation from 0.5 mg·L^-1 to 50.0 mg·L^-1(r^2=0.999,n=7).Method recovery of LZD was greater than or equal to 91.69%.Extraction recovery of LZD was greater than or equal to 81.50%.RSDs of intra-day and inter-day precision were less than or equal to 10.30%and 11.50%,respectively.RSD of long term stability,repeated freezethaw stability were less than or equal to 13.52%and 5.36%respectively.RSD of room-temperature stability(samples were placed at room temperature for 12 h)was less than or equal to 10.76%.The patients'plasma concentrations of LZD were within 0.73– 6.30 mg·L^-1.Conclusion:A rapid,simple,reliable HPLC method was established in our study for clinical therapeutic drug monitoring(TDM)of LZD.
