球囊损伤血管内膜组织CD36和Prdx-2蛋白表达

摘　　要：目的观察球囊损伤血管内膜的增生情况及其内CD36和过氧化物酶基因-2(Prdx-2)蛋白的表达。方法将32只雄性Wistar大鼠随机分为假手术对照组和球囊损伤手术组,每组16只。球囊损伤手术组用球囊损伤大鼠主动脉内膜。分别于术后14、28d取两组大鼠主动脉血管壁,行苏木精-伊红染色后测量血管壁内膜和中膜厚度,并用免疫组织化学方法检测血管内膜中CD36和Prdx-2蛋白的表达。结果球囊损伤术后14、28d,大鼠血管壁内膜及中膜显著增生。球囊损伤后血管内膜中CD36蛋白表达水平增高,与血管内膜厚度呈正相关(r=0.662,P<0.01);Prdx-2蛋白表达水平降低,与血管内膜厚度呈负相关(r=-0.516,P<0.05)。结论血管内皮损伤修复过程中存在内膜增生,CD36蛋白表达上调及Prdx-2蛋白表达下调可能与血管平滑肌细胞移行、增生及损伤后再狭窄的发生有关。Objective To investigate intimal hyperplasia after balloon injury and the expression of CD36and Prdx-2in aortic intima.Methods A total of32male Wistar rats were randomly divided into sham-operation control group and balloon injury group,with16rats in each group.In the balloon injury group,the aortic intima was injured by a balloon.The aortic wall was collected on days14and28after surgery;hematoxylin and eosin staining was performed to measure intima-media thickness,and immunohistochemistry was used to measure the expression of CD36and Prdx-2proteins in the aortic intima.Results There was significant intimal-medial hyperplasia on days14and28after surgery.After balloon injury,there was an increase in the expression of CD36protein in the aortic intima,which was positively correlated with intimal thickness(r=0.662,P<0.01),and there was a reduction in the expression of Prdx-2protein,which was negatively correlated with intimal thickness(r=-0.516,P<0.05).[WTHX]ConclusionIntimal hyperplasia is observed during the repair of vascular endothelial injury.Upregulation of CD36protein and downregulation of Prdx-2protein may be associated with the migration and proliferation of vascular smooth muscle cells and res-tenosis after injury.
