摘 要:目的探讨将氟代脱氧葡萄糖(^(18)F-FDG)和6-^(18)F-L-多巴(^(18)F-DOPA)配制成[^(18)F-FDG+^(18)F-DOPA]复方制剂进行正电子发射及计算机断层扫描成像系统(PET-CT)显像的效果。方法分别合成^(18)F-FDG及^(18)F-DOPA,并配制成放射性活度比^(18)F-FDG∶^(18)F-DOPA分别为4∶6、5∶5和6∶4的[^(18)F-FDG+^(18)F-DOPA]复方制剂。分别用^(18)F-FDG、^(18)F-DOPA及3种配比的复方制剂对Wistar大鼠进行PET-CT显像。结果全自动化合成^(18)F-FDG和^(18)F-DOPA,产品质量均达到动物和人体PET/CT显像要求。PET-CT显像均可见双侧纹状体对称性放射性摄取。纹状体与脑皮质比值分别为:1.08(^(18)F-FDG)、1.35(^(18)F-DOPA)、1.33(^(18)F-FDG∶^(18)F-DOPA=5∶5)、1.44(^(18)F-FDG∶^(18)F-DOPA=4∶6)、1.16(^(18)F-FDG∶^(18)F-DOPA=6∶4)。结论从纹状体与脑皮质比值及PET-CT显像效果可以看出[^(18)F-FDG+^(18)F-DOPA]复方制剂中^(18)F-FDG∶^(18)F-DOPA为4∶6的制剂最为理想。该实验结果为进一步开展模型鼠显像提供了基础,[^(18)F-FDG+^(18)F-DOPA]复方制剂的应用价值有待进一步研究。Objective To synthesize [18 F-FDG + 18 F-DOPA] compound by using 18 F-FDG and18 F-DOPA and evaluate the positron emission tomography/computed tomography(PET-CT) imaging effect ofthe synthesized compound. Methods 18 F-FDG and 18 F-DOPA were separately synthesized.The compound[18 F-FDG + 18 F-DOPA] tracers with 18 F-FDG to 18 F-DOPA radioactivity ratios of 4∶6, 5∶5 and 6∶4 wereprepared. The PET-CT images of Wistar rats were also performed with 18 F-FDG, 18 F-DOPA and three compoundpreparations above. Results 18 F-FDG and 18 F-DOPA were automated synthesized. The product quality metthe animal and human PET-CT imaging requirements. PET-CT images revealed symmetrical radioactive uptakeof bilateral striatum. The ratios of striatum and cerebral cortex were 1.08(18 F-FDG), 1.35(18 F-DOPA), 1.33(18 F-FDG∶18 F-DOPA = 5∶5), 1.44(18 F-FDG∶18 F-DOPA = 4∶6) and 1.16(18 F-FDG∶18 F-DOPA = 6∶4) respectively. Conclusions The compound [18 F-FDG + 18 F-DOPA] tracer with the radioactivity ratio of 4∶6 yields the highest imaging quality according to the ratio of striatum and cerebral cortex and PET-CT imagingeffect. The experimental results provide a basis for subsequent establishment of PET-CT mouse models. Theapplication value of [18 F-FDG + 18 F-DOPA] compound remains to be investigated.
