摘 要:目的:观察补肾方、补肾健脾方、益气健脾化痰祛瘀方对快速老化模型小鼠心肌葡萄糖转运体蛋白的影响,探讨中医不同治法心肌保护作用的机制。方法:选用快速老化模型小鼠,分为对照组、模型组、补肾方组、补肾健脾方组、益气健脾化瘀祛痰方组。连续灌胃给药3个月,采用生化分析仪检测血清TC和LDL-C,采用ELISA法检测血清IL-6水平,采用HE染色法观察胸主动脉和心肌组织形态,采用油红O染色法观察胸主动脉脂质面积,采用Masson染色法观察胸主动脉胶原纤维的变化,采用Westernblot法检测心肌组织GLUT-1和VCAM-1蛋白表达。结果:与对照组比较,模型组血清TC、LDL-C和IL-6水平明显升高(P<0.05,P<0.01),胸主动脉和心肌组织病理形态发生改变,心肌组织GLUT-1蛋白表达降低和VCAM-1蛋白表达显著升高;与模型组比较,给药组血清TC和LDL-C水平降低(P<0.01),改善胸主动脉和心肌组织的病理变化,益气健脾化痰祛瘀方组血清IL-6水平降低(P<0.05),提高心肌组织GLUT-1蛋白表达和抑制心肌组织VCAM-1蛋白表达。结论:补肾方、补肾健脾方、益气健脾化瘀祛痰方对快速老化模型小鼠具有代谢调节、心肌保护、血管保护三重作用,改善衰老所致的心肌能量代谢障碍、血管损伤,炎症反应,其作用机制可能与其干预心肌葡萄糖转运体蛋白有关,以益气健脾化痰祛瘀方效果较为明显。Objective:To study the effect of different formulae on glucose transporter of heart in SAMP6 mice and investigate its mechanism.Methods:Mice were assigned into 5 groups:the control group(SAMR1 group),the model group(SAMP6 group),Bushen Recipe group,Bushen Jianpi Recipe group,Yiqi Jianpi Huayu Qutan Recipe group.At the end of the three months,the total cholesterol(TC),low density lipoprotein(LDL-C)and interleukin-6(IL-6)in serum were detected.The aortic and heart were harvested for HE,oil-red-O and Masson staining.The protein expressions of glucose transporter 1(GLUT-1)and vascular cell adhesion molecule-1(VCAM-1)in the heart were measured by Western blot.Results:Comparing with the model group,different formulae can improve the histology morphology of aortic and heart.It also can significantly reduce the levels of TC and LDL-C in the serum(P<0.01).Yiqi Jianpi Huayu Qutan Recipe can down-regulate IL-6 in the serum(P<0.05).It up-regulated GLUT-1 and down-regulated VCAM-1 in heart.Conclusion:Different formulae can regulate metabolic disturbances,impairment in heart and the inflammatory factors.
