黄芪丹参配伍对大鼠骨髓源EPCs功能的影响

摘　　要：目的:观察黄芪丹参配伍对体外培养大鼠骨髓源内皮祖细胞(EPCs)功能的影响。方法:采用密度梯度离心法配合差速贴壁法分离培养大鼠骨髓源EPCs,用MTT法、Transwell小室法、黏附能力实验和Matrigel胶法检测黄芪、丹参、当归等配伍对体外培养大鼠骨髓源EPCs增殖、迁移、黏附和形成管状结构功能的影响。结果:单味黄芪、丹参、当归以及黄芪丹参配伍、黄芪当归配伍均具有改善EPCs生物学功能的作用,其中以黄芪丹参配伍组改善EPCs功能的作用最显著,与黄芪组、丹参组、当归组和空白对照组相比,存在显著性差异(P<0.05)。结论:黄芪丹参配伍较黄芪、丹参、当归单味中药能更好地提高EPCs的增殖、迁移、黏附和形成管状结构的能力,黄芪和丹参中的某些活性成分在提高EPCs生物学功能的过程中可能具有协同作用,亦提示益气活血治法可能对改善EPCs的某些功能具有一定的作用。Objective:To observe the effects of astragalus and salvia compatibility serum on the rats bone marrow-derived endothelial progenitor cells(EPCs).Methods:EPCs were separated from the rat marrow by density gradient centrifugation and purified by differential adhesion.MTT assay,transwell chamber method,cell adhesion test and matrigel method were used to evaluate EPCs'proliferation,transferring,adhesion and tubular structure formation ability respectively.Results:Astragalus,salvia,angelica,astragalus and salvia compatibility,astragalus and angelica compatibility serum are all can improve the biological functions of EPCs,and the strongest one is astragalus and salvia compatibility serum group when compared with Astragalus,salvia and angelica serum groups and blank serum group,and the difference had statistical significance.Conclusion:The effect of enhancing EPCs'biological functions of astragalus and salvia compatibility serum is more significant than that of astragalus,salvia,angelica and astragalus and angelica compatibility serum.Some active ingredients in astragalus and salvia may have synergism to increase the biological function of EPCs.Supplementing Qi and activating blood circulation method can significantly improve the biological function of EPCs.
