PCPA失眠大鼠大脑皮质星形胶质细胞GFAP和P2X7受体表达的变化及酸枣仁汤的干预作用

摘　　要：目的:研究PCPA诱导大鼠失眠后大脑皮质星形胶质细胞GFAP和P2X7R表达的变化及SZRD的干预作用。方法:采用连续3d腹腔注射PCPA的方法复制失眠模型,大鼠随机分为对照组,模型组,中药治疗组和中药对照组,中药治疗组和中药对照组给予SZRD灌胃7d。各实验组分别在第4、5、6、8、10天取大鼠大脑皮质前额叶,采用免疫荧光双染法检测各组大鼠不同时间大脑皮质GFAP和P2X7R蛋白表达,用实时荧光定量PCR法测定各组大鼠不同时间大脑皮质P2X7R基因mRNA表达。结果:与对照组比较,模型组大鼠不同时间大脑皮质GFAP、P2X7R蛋白及mRNA表达均明显上调(P<0.01);与模型组比较,第5、6、8、10天中药治疗组GFAP表达逐渐下降(P<0.05,P<0.01),第6、8、10天中药治疗组P2X7R蛋白表达逐渐降低(P<0.05,P<0.01),第8、10天中药治疗组P2X7RmRNA表达水平逐渐下调(P<0.05,P<0.01)。结论:SZRD干预PCPA所致失眠的机制可能与调控大鼠大脑皮质星形胶质细胞GFAP和P2X7R的表达有关。Objective:To investigate the changes of GFAP and P2X7R expression in astrocytes of cerebral cortex after PCPA induced insomnia rats and the intervention effect of Suanzaoren Decoction(SZRD).Methods:The insomnia model was reproduced by intraperitoneal injection of PCPA for 3 days.Rats were randomly divided into control group,model group,traditional Chinese medicine treatment group and traditional Chinese medicine control group.The traditional Chinese medicine treatment and control groups were given SZRD for 7 days.The prefrontal cortex of rats was taken out at the 4th,5th,6 th,8 th and 10th day in each experimental group.The expressions of GFAP and P2X7R protein in the cerebral cortex were examined using immunofluorescence double staining,and the expression of P2X7R mRNA in the cerebral cortex was examined using real-time quantitative PCR.Results:Model group’s rats showed significantly higher expression levels of GFAP,P2X7R protein and mRNA in the cerebral cortex than control animals at different time points.Compared with the model group,GFAP expression decreased gradually on the 5th,6th and 8th day,P2X7R protein expression of the treatment group decreased gradually on the 6th and 8th day,and P2X7R mRNA expression of the treatment group was gradually down-regulated.Conclusion:The mechanism of SZRD intervention in PCPA induced insomnia may be related to the regulation of GFAP and P2X7R expression on astrocytes in the cerebral cortex of rats.
