摘 要:本文将干法人工授精的施氏鲟Acipenser schrenckii卵用滑石粉脱粘后在水流量600L/h水族箱中,添加0mg/L(对照)、1.0mg/L、2.5mg/L、5.0mg/L和10mg/L谷氨酰胺(Gln),测定胚胎发育总孵化率、畸形率、谷胱甘肽(GSH)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)的活力和丙二醛(MDA)的含量,以探索提高施氏鲟胚胎孵化抗氧化损伤能力和孵化效果的方法。结果显示:添加1.0~10.0mg/L Gln能缩短施氏鲟胚胎发育时间、提高总孵化率、降低畸形率、促进各发育期GSH合成、提高SOD、CAT酶活力和降低MDA含量,其中5.0 mg/L变化幅度最大,是施氏鲟胚胎孵化期间的最佳添加量。研究同时表明原肠期胚胎GSH含量、SOD酶活力、CAT酶活力和MDA含量变化幅度较小,而心跳期和出膜前期变化幅度较大。Used dry artificial insemination Acipenser schrenckii eggs with talcum powder after deboning in water flow 600 L/h aquatic animals, to add 0 mg/L(control), 1.0 mg/L, 2.5 mg/L, 1.0 mg/L and 10 mg/L glutamine(Gln), measured the total incubation rate,deformity rate, glutathione(GSH) content,superoxide dismutase(SOD) activity, catalase(CAT) activity, and malondialdehyde(MDA)content, to research on improving the Acipenser schrenckii embryo incubation oxidative damage resistance and the effect of incubation method. The results showed that add 1.0 mg/L ~ 10.0 mg/L Gln can shorten the embryonic development time increase the total incubation rate, reduce the deformity rate, promote the synthesis of GSH and increase the activity of SOD and CAT enzyme in each developmental stage. Among them, 5.0 mg/L has the largest change, which is the best addition of Gln to the embryo hatching of Schneider.The study also showed that the changes of GSH content, SOD enzyme activity, CAT enzyme activity and MDA content were small,while the changes of heart jump period and premembrane were large.
