摘 要:[目的]对黑骨藤不同极性部位进行急性毒性试验及抗氧化研究,确定其临床用药安全性及活性部位。[方法]急性经口毒性试验采用SD(180~220 g)大鼠,连续灌胃给药7 d(9.75 g/kg)后,测定血清中的肝功能和肾功能等相关指标,观察其肝脏、肾脏毒性反应,采用ABTS(agreement of basic telecommunications services)法测定肝脏中的总抗氧化能力。[结果]黑骨藤不同极性部位给药7 d后,大鼠未出现死亡,石油醚部位、氯仿部位、醋酸乙酯部位、正丁醇部位组对肝脏和肾脏生化指标均无显著性影响(P>0.05),水部位组大鼠血清中的ALP和BUN显著升高(P<0.05)。[结论]黑骨藤的不同极性部位中的石油醚部位、氯仿部位、醋酸乙酯部位、正丁醇部位未出现毒性反应,水部位中的ALP和BUN升高,可能表现出微低毒性。氯仿部位和正丁醇部位表现出较强的抗氧化能力。[Objective]To determine the safety of clinical medication and active fractions screening of Perploca forrestii Schltr.,we conducted acute toxicity experiment and antioxidative study of different polarity extractions from Perploca forrestii.[Methods]SD rats(180-220 g)used to study acute oral toxicity test,was given different polarity extractions from Perploca forrestii.9.75 g/kg by gavage for 7 days,to determinate the relevant indicators of liver function and renal function etc in serum,and to observe the toxicity of liver and kidney.ABTS(agreement of basic telecommunications services)used to determinate of total antioxidant capacity in the liver.[Result]The rats were given drugs by gavage for 7 days,and did not die,there was no significant effect on liver and kidney biochemical indexes in petroleum ether,chloroform,ethyl acetate and n-butanol group(P>0.05),the ALP and BUN were significantly increased in the serum of the rat of water group(P<0.05).[Conclusion]There was no toxic reaction in the petroleum ether,chloroform,ethyl acetate and n-butanol group,elevation of ALP and BUN in the water group may show microtoxicity.The oxidation resistance of chloroform and n-butanol group was strong.
