摘 要:目的探讨人参提取物对Ⅰ型干扰素及下游基因表达的调节作用。方法培养THP-1细胞并加入人参提取物后,ELISA检测干扰素-α、干扰素-β分泌水平,Real-time PCR检测BST-2、APOBEC3G表达。结果与空白组比较,人参提取物组干扰素-α分泌水平显著提高(P<0. 01),但对干扰素-β无明显影响(P>0. 05);与对照siRNA组比较,该组BST-2、APOBEC3G表达显著提高(P<0. 01),并与干扰素-α诱导呈正相关。结论人参提取物可通过Toll样受体途径诱导单核细胞分泌干扰素-α,从而提高抗病毒因子表达。AIM To explore the regulatory effects of Ginseng Radix et Rhizoma extract on expressions of type I interferons and downstream genes. METHODS After the addition of Ginseng Radix et Rhizoma extract into cultured THP-1 cells,the detection of interferon-α,interferon-β secretion levels and BST-2,APOBEC3G expressions by ELISA and Real-time PCR were practiced,respectively. RESULTS Ginseng Radix et Rhizoma extract group exhibited significantly increased interferon-α secretion level ( P<0. 01) without obvious effect on that of interferon-β( P>0. 05) when compared with the blank group; and it also demonstrated markedly increased BST-2,APOBEC3G expressions ( P<0. 01),showing a positive correlation with interferon-α induction when compared with siRNA control group. CONCLUSION Ginseng Radix et Rhizoma extract can induce interferon-α secretion of monocytes via Toll-like receptor pathway and thus improve the antiviral factors expressions.
