减味寿胎丸对药源性卵巢功能损伤大鼠的干预机理初探

摘　　要：目的:本研究以雷公藤多苷致卵巢功能损伤模型为载体,运用去阿胶减味寿胎丸初步探索其对卵巢功能损伤的干预及其机理,以期指导临床治疗。方法:选择有正常动情周期SPF级SD大鼠随机分成:正常组、模型组、补佳乐组、减味寿胎丸高及低剂量组,每组10只,共50只。模型组、补佳乐组及中药高、低剂量组分别于上午9时灌服同剂量TW75mg/kg,下午3时按分组情况分别灌服同等剂量生理盐水、补佳乐或中药;正常组则同时灌服同剂量生理盐水,连续30日。实验结束后,均选择1～5天非动情期处死、取材。结果:减味寿胎丸可使部分大鼠(约半数)动情周期恢复;可使大鼠FSH,FSH/LH明显降低(P<0.05);血E2水平升高,虽P>0.05; AMH、INHB明显升高,卵巢指数明显升高(P<0.05);可使生长卵泡增加,但P>0.05。可使生长卵泡增多、颗粒细胞层数增加、间质纤维化改善。结论:减味寿胎丸可能是通过提高卵巢对促性腺激素的反应性和调节卵巢内分泌激素的水平,改善卵巢血液供应及组织形态,从而恢复和改善卵巢功能。Objective:The ovarian function damage model induced by Tripterygium wilfordii polysaccharide was used as the carrier to explore the intervention and mechanism of the pill to reduce the function of ovarian function,so as to guide the clinical treatment.Methods:the rats with normal estrous cycle SPF grade SD rats were randomLy divided into normal group,model group,bujiale group,high and low dose group,with 10 rats in each group,with a total of 50 rats.The model group,bujiale and high and low dose group of Chinese medicine were given the same dose of TW 75mg/kg at 9 am,and the same dosage of normal saline,GIC or Chinese medicine were given respectively at 3 pm and the normal group was given the same dose of normal saline at the same time,all for 30 days.At the end of the experiment,1~5 days of non estrus were selected.Results:the estrous cycle of some rats could be recovered and the FSH and FSH/LH decreased significantly(P<0.05),the level of blood E2 increased,although P>0.05,AMH and INHB increased significantly(P<0.05),and the growth follicle increased,but P>0.05.It can increase the number of growing follicles,increase the number of granular cells,and improve interstitial fibrosis.Conclusion:It is possible to improve the ovarian function by improving the reactivity of ovary to gonadotropin and regulating the level of ovarian endocrine hormone,improving the ovarian blood supply and tissue morphology,so as to restore and improve the ovarian function.
