摘 要:为探讨蒲公英水提物对猪源性大肠杆菌刺激小鼠肝脏损伤的保护作用,建立实验动物模型,分别设立蒲公英水提物低、中、高剂量组、空白、模型组。试验周期为7d,第8天取血和肝脏,测定各组小鼠肝组织中GSH、SOD活性及血清中炎性细胞因子TNF-α、IL-6、IL-1β、IFN-γ和IL-4的表达量。结果表明:各剂量的蒲公英水提物均能一定程度地提高应激性肝损伤小鼠肝组织中GSH和SOD的活性,降低炎症反应所致血清炎性因子TNF-α、IL-6、IL-1β、IFN-γ和IL-4的表达量。表明蒲公英水提物通过降低血清炎性因子的表达量、升高肝组织GSH和SOD活性以降低炎症反应,提高肝脏抗氧化能力,从而对肝脏起到保护作用。To investigate the protective effects of dandelion extract against hepatic injury induced by Escherichia coli from swine,the model of liver injury was established in mice,the low,medium and high dose groups of dandelion extract,blank group,model group were set up.The experiment lasted 7days,the mice were sacrificed on the 8th day.The activity of GSH and SOD in liver and the expression level of inflammatory cytokines TNF-α,IL-6,IL-1β,IFN-γand IL-4in sera were measured and the pathological changes of liver were observed under microscope.The results showed that the crude extract of dandelion could increase GSH and SOD activities and decrease the expression levels of serum inflammatory cytokines TNF-α,IL-6,IL-1β,IFN-γand IL-4.It suggests that dandelion extract can protect against liver injury of mice by reducing the expression of serum inflammatory cytokines,increasing the activity of hepatic GSH and SOD.
