摘 要:目的研究通补络脉法对特发性肺纤维化(IPF)模型大鼠下丘脑五羟色胺(5-hydroxytryptamine,5-HT)及肺α-平滑肌动蛋白(α-smooth muscle action,α-SMA)含量的影响机制。方法将48只雄性Wistar大鼠常规饲养1周后,随机分为3组:空白对照组、模型对照组和通补络脉组。空白对照组气管内注入生理盐水,模型对照组、通补络脉组均用气管内注入博莱霉素复制肺纤维化模型,各组均于造模次日起开始灌胃,分别于第14天、第28天随机选取8只处死后取材。通过MASSON染色观察病理组织变化,确定肺纤维化程度;称取大鼠肺湿重及体重,计算肺系数; ELISA法测定大鼠组织5-HT及α-SMA含量。结果模型对照组肺组织Masson染色病理切片的特点是肺泡炎症渐变致纤维化的动态改变;与空白对照组比较,模型对照组大鼠肺系数、下丘脑5-HT以及肺组织α-SMA含量显著升高(P <0. 05);与模型对照组比较,通补络脉组大鼠肺系数、下丘脑5-HT以及肺组织α-SMA含量明显降低(P <0. 05)。结论通补络脉法可以通过干预IPF大鼠肺组织α-SMA的表达以及降低下丘脑5-HT的含量,以减轻肺纤维化程度,其内在机制可能与"神经—内分泌—免疫(NEI)"网络调节有关。Objective To explore the mechanism on the influences of tongbu luomai method on 5-hydroxytryptamine(5-HT) of the hypothalamus and α-smooth muscle action(α-SMA) of the lung tissue in the rat models of idiopathic pulmonary fibrosis(IPF). Methods A total of 48 Wistar male rats were randomized into three groups after the regular feeding for 1 week,i. e. a blank control group,a model control group and a tongbu luomai group. In the blank control group,the normal saline was infused into the trachea. In the model control group and the tongbu luomai group,bleomycin was injected into the trachea to duplicate IPF model. The gavage started on the second day of modeling in each group. Separately,on the 14 th and 28 th days,8 rats were selected randomly and sacrificed for sample collection. MASSON staining was adopted to observe the pathological changes in the tissues and determine the severity of pulmonary fibrosis.The lung wet weight and body mass were measured and the lung coefficient was calculated. ELISA method was applied to determine the levels of 5-HT and α-SMA. SPSS 19. 0 software was provided for the statistical analysis. Results In the model control group,the characteristics of the pathological slice in the lung tissue with Masson staining indicated the dynamic change of the gradual fibrosis of alveolar inflammation. Compared with the blank control group,the lung coefficient and the levels of 5-HT of the hypothalamus and α-SMA of the lung tissue in the rats were increased significantly in the model control group(P < 0. 05). Compared with model control group,the lung coefficient and the levels of 5-HT of the hypothalamus and α-SMA of the lung tissue in the rats were increased significantly in the tongbu luomai group(P <0. 05). Conclusion Tongbu luomai method alleviates pulmonary fibrosis through changing the expression of α-SMA of the lung tissue and reducing the level of 50 HT in the hypothalamus in the rats. The effect mechanism is probably related to the regulation of nerve-endocrine-immunity network.
