摘 要:为研究露地环境中栒子试管苗的瓶内生根培养和移栽技术,在天津农学院地被植物园的露地环境中,于4月下旬采用二层遮阳网、三层遮阳网及树荫3种遮阴方式进行栒子试管苗瓶内生根培养,并采用2种覆膜保湿方式和2个时期进行栒子试管苗的移栽试验。结果表明:3种遮阴方式下栒子试管苗瓶内生根培养的效果无明显差异,可获得89.7%~95.9%的生根率,试管苗生长正常。二层遮阳网遮阴下培养的栒子试管苗气孔关闭功能明显强于另外两种遮阴方式。采用完全覆膜和顶部开口覆膜的保湿方法,栒子试管苗移栽成活率无显著差异,6月1日和6月27日移栽的栒子试管苗的成活率也无显著差异,移栽成活率均达到97.5%~100.0%。本结果可为降低栒子试管苗成本及促进组织培养快速繁殖技术的生产应用提供依据。The aim of this study was to research on in vitro rooting and transplanting of Cotoneaster hjelmqvistii plantlets in vitro in field environment.Three shading methods(shading with two layers of sunshade net,three layers of sunshade net or tree shade)were used for in vitro rooting of C.hjelmqvistii plantlets in field environment in last ten days of April,and transplanting experiments were carried out with two preserve moisture cover methods in two periods.The results showed that there was no significant difference in the effect of root culture among the three shading methods,and the rooting rate of 89.7%-95.9%were obtained meanwhile the plantlets grew normally.The closure function of the stomata in treatment of shading with two layers of sunshade net was obviously stronger than that of the other two shading methods.There was no significant difference in the survival rate of the plantlet transplanted between both preserve moisture cover methods of the fully cover and the cover with top opening;the survival rate of transplanted plantlets in June 1st and June 27th had no significant difference,and the survival rate was all up to 97.5%-100.0%.The results can provide a basis for reducing the cost of C.hjelmqvistii plantlets in vitro and promoting the tissue culture rapid propagation technique application to reproduction.
