中医病证相符的大鼠疮疡模型的建立及其血浆、尿液代谢组学研究

摘　　要：目的:观察胶带表皮剥脱加金葡菌菌液注射方法致大鼠背部疮疡模型的造模效果。方法:20只SD大鼠分为正常组和模型组各10只,模型组麻醉后背部脱毛,以胶带粘扯背部软毛脱表皮,滴注菌液待其自然苏醒,以后连续3d于其皮下注射菌液,正常组以生理盐水代替菌液。至第4天取局部组织进行HE染色和革兰氏染色,收集血浆及尿液备检测。通过观察正常组大鼠与模型组大鼠全身及局部疮面情况、病理标本,判断大鼠疮疡模型建模成功。利用代谢组学分析方法,对正常组与模型组大鼠的血浆与尿液进行代谢轮廓分析。结果:模型大鼠表现出阳证疮疡中期成脓的证候特点,病理切片观察显示造模成功,代谢组学检测显示实验模型大鼠血浆、尿液的图谱与正常组大鼠的图谱存在一定的差异,PCA图可以看出正常组与模型组大鼠血浆、尿液代谢谱分为两类,两者明显区分。OPLS-DA图可以看出正常与模型大鼠的血浆、尿液生物样品呈现聚拢状态,且区分明显。结论:模型大鼠表现出中医"阳证"证型特点,胶带表皮剥脱菌液滴注加皮下菌液注射造模方法可制作出符合实验需要的体表感染疮疡模型(中期)。Objective:To observe the effect of pyocutaneous model induced by the method of adhesive tape stripping and Staphylococcus aureus liquid injection.Methods:20 SD rats were randomly divided into the normal group(n=10)and the model group(n=10).The rats in the model group were removed the back hair after anesthesia,and stripped the back fur skin with adhesive tape.Bacteria liquid was infused followed by the injection of bacteria for continuous 3 days subcutaneously and the normal group was given normal saline.The fourth day local tissues were stained with HE and Gram staining,at the same time plasma and urine were collected for testing.The local wound and pathological specimens were evaluated to determine whether the pyocutaneous model was successful.The metabolic profiles of plasma and urine were analyzed by UHPLC-QTOF/MS method in the normal group and in the model group.Results:The model rats showed Yang syndrome of pyocutaneous with pus formation in the mid-term.Pathological observation showed the modeling was successful.Metabolic profiling showed that there were some differences in the plasma and urine between the model group and the normal group.PCA chart showed that metabolic profiles of plasma and urine were divided into two categories in the two groups.OPLS-DA chart showed plasma urine biological samples in cage state and distinguished significantly in the model group and in the normal group.Conclusion:Model rats showed characteristic of Yang syndrome,the model induced by adhesive tape stripping and Staphylococcus aureus liquid injection consisted with pyocutaneous model of skin infection of mid-term.
