摘 要:目的:观察姜黄素(curcumin,Cur)对高糖腹膜透液(PDF)作用下人腹膜间皮细胞株(HMrSV5细胞)转分化(EMT)及转化生长因子β_1(TGF-β_1)分泌的影响。方法:HMrSV5细胞常规培养、传代后用于实验。随机分组为,正常对照组、不同浓度高糖PDF(1. 5%、2. 5%、4. 25%)组和4. 25%PDF+不同浓度姜黄素(20、40、80μmol/L)组; Real time RT-PCR检测细胞α-SMA和E-cadherin的mRNA表达水平; Western印迹方法检测细胞α-SMA、E-cadherin的蛋白表达水平。ELISA法检测细胞培养上清液中TGF-β_1的含量。结果:与正常对照组相比,高糖PDF处理后,HMrSV5细胞中α-SMA表达明显上调,E-cadherin表达明显下调,并呈浓度依赖性(均P <0. 05)。高糖可促进HMrSV5细胞分泌TGF-β_1(P <0. 05)。姜黄素可抑制高糖PDF诱导的HMrSV5细胞α-SMA表达上调、E-cadherin表达下调及TGF-β_1分泌增加,并呈浓度依赖性(均P <0. 05)。结论:高糖PDF可诱导人腹膜间皮细胞的EMT效应。姜黄素以浓度依赖性效应抑制人腹膜间皮细胞的EMT,这种效应可能与姜黄素抑制TGF-β_1分泌有关。Objective: To explore the effect of Cur on the epithelial-mesenchymal transition (EMT) and the excretion of transforming growth factor-β1 (TGF-β1) in human peritoneal mesothelial cells (HPMCs) induced by high glucose-based peritoneal dialysis fluids(PDF). Methods: Cultured human peritoneal mesothelial cells (HMrSV5) were divided into three groups: normal control group, high glucose-based PDF(1.5%,2.5%,4.25%) group and high glucose-based PDF(4.25%) plus low-, medium-and high-concentration Cur (20,40,80 μmol/L ) group respectively. Real time RT-PCR was used to detected the expression of α-smooth muscle actin (α-SMA) and E-cadherin mRNAs in HMrSV5 cells. Expression levels of α-smooth muscle actin (α-SMA) and E-cad herin in protein level were detected by Western blotting and the concentration of TGF-β1 in the culture supernatant was determined by ELISA. Results: Compared with normal control group, High glucose-based PDF treated HMrSV5 cells exhibited a dose-dependent increase in the expression of α-SMA, and decrease in the expression of E-cadherin both at protein and mRNA levels ( P <0.05). PDF also stimulated HMrSV5 cells to secrete TGF-β1. Treated with Cur, high glucose-based PDF induced α-SMA expression and TGF-β 1 production were reduced, E-cadherin expression was increased. Conclusion: Curcumin can inhibit high glucose-based PDF induced EMT of human peritoneal mesothelial cells by down?regulating the expression of TGF-β1.
