摘 要:目的探讨麦冬皂苷D'对大鼠心肌细胞H9c2的细胞毒性作用。方法麦冬皂苷D'0. 1,0. 5,5,10μmol/L作用于H9c2细胞24 h,MTS法检测细胞毒性,流式细胞仪检测细胞凋亡率、细胞周期和线粒体膜电位。结果不同浓度麦冬皂苷D'作用于H9c2细胞24 h,5μmol/L及以上浓度的药物处理表现出明显的细胞毒性;流式细胞仪检测显示,5μmol/L及以上浓度的麦冬皂苷D'可诱导H9c2细胞凋亡(P <0. 05),将心肌细胞阻滞于G_0/G_1期(P <0. 01),降低线粒体膜电位(P <0. 05)。结论较高浓度的麦冬皂苷D'对H9c2心肌细胞有明显的细胞毒性作用。Objective To investigate the cytotoxicity of Ophiopogonin D'(OPD') on rat cardiomyocytes H9c2 cells. Methods H9c2 cells were treated with 0. 1, 0. 5, 5 and 10 μmol/L OPD' for 24 h. The cytotoxicity was measured by MTS assay, and the apoptosis rate, cell cycle, and mitochondrial membrane potential were measured by flow cytometry. Results Different concentration of OPD' were treated on H9c2 cells for 24 h, 5μmol/L and above concentrations of OPD' showed significant cytotoxicity. Flow cytometry results showed that 5 μmol/L and above concentrations of OPD' could induce apoptosis in H9c2 cells( P < 0. 05), block H9c2 cells in Go/Gi phase (P < 0. 01), and decrease mitochondrial membrane potential(P < 0. 05). Conclusion The higher concentration of OPD' has significant cytotoxic effects on H9c2 cells.
