摘 要:试验旨在研究CAPN1基因在早胜牛群体中的遗传变异,为进一步寻找与早胜牛肉质性状相关的分子标记奠定基础。采用DNA混合池测序法对早胜牛320个样本CAPN1基因所有外显子的SNPs位点进行筛选,同时进行序列分析,并利用测序图中SNP位点等位基因峰高的比值估算各等位基因的频率。结果表明:在早胜牛CAPN1基因外显子区共筛选到7个多态性位点G3717A、A3854G、C5709G、C6004T、G11058A、G15299A和G15682A,其中G3717A和G15299A为同义突变,其余5个SNPs位点为错义突变。该研究结果将为进一步研究早胜牛肉质性状相关候选基因提供理论依据。The aim of this study was to investigate the genetic variation of the CAPN1 gene in Zaosheng cattle population,and to lay the foundation for further searching for molecular markers associated with beef quality traits of Zaosheng cattle.In this study,DNA pool sequencing was used to screen the SNPs of all exons of the CAPN1 gene in the Zaosheng cattle of which 320 samples,and sequence analysis was performed.The frequency of alleles was estimated by using the ratio of the alleles peak height of the SNP site in the sequencing diagram.The results showed that seven polymorphic loci were screened in exons of CAPN1 gene of Zaosheng cattle:G3717A,A3854G,C5709G,C6004T,G11058A,G15299A and G15682A,of which G3717A and G15299A were silent mutations,and the remaining five SNPs were missense mutation.The results of this study will provide a theoretical basis for the study of candidate genes related to beef quality traits of Zaosheng cattle.
