摘  要:为了促进药食两用植物忧遁草资源的开发利用,将忧遁草无菌茎段置于液体培养基中,采用正交设计法优化植物生长调节剂组合、基本培养基类型、NO_3^-/NH_4^+值、PO_4^(3-)和蔗糖质量浓度,以繁殖系数为指标,建立其悬浮高效繁殖的工艺。结果表明,改良的1/2MS培养基(20 mmol/L KNO_3+5 mmol/L NH_4NO_3+1. 5 mmol/L KH_2PO_4)+蔗糖20 g/L+IBA 1. 0 mg/L+6-BA 0.5 mg/L+NAA0. 8 mg/L+KT 0. 2 mg/L(pH值6. 0)是最适合忧遁草茎段悬浮培养的配方,其无菌苗根数为4. 09个,根长为5. 47 cm,茎高为11. 79 cm,植株形态为叶片展开充分,叶色深绿,植株健壮。In order to promote the development and utilization of dietetic Chinese herbs-Clinacanthus nutans,stem segments from sterile seedlings of Clinacanthus nutans were placed into liquid culture.Using propagation coefficient as indicator and orthogonal design as experiment mothods,the efficient protocol towards suspension culture was established through optimum combination of phytohormone,basic media,ratio of NO3^-/NH4^+,concentrations of PO4^3-and sucrose.The results showed that the combination of modified 1/2MS+20 mmol/L KNO 3+5 mmol/L NH4NO3+1.5 mmol/L KH 2PO 4+20 g/L sucrose+IBA 1.0 mg/L+6-BA 0.5 mg/L+NAA 0.8 mg/L+KT 0.2 mg/L(pH 6.0)was the most suitable formula for propagation of Clinacanthus nutans seedlings under liquid culture conditions.The obtained plants had expanded and dark green leaves,and vigorous stems with mean root number of 4.09,root length of 5.47 cm,and plant height of 11.79 cm.
