摘  要:为了给猕猴桃单倍体育种提供适宜发育时期的小孢子,以‘东红'猕猴桃雄株花蕾为材料,用醋酸洋红压片法制片,在生物数码显微镜下观察花粉发育情况,并探讨花蕾形态特征与花粉发育时期的相互关系。结果表明,Ⅰ期、Ⅱ期花蕾的瓣萼比分别为1. 59和1. 67,小孢子发育时期大多处于单核期,所占比例分别为84. 4%和68. 6%; III期、IV期花蕾的瓣萼比分别为1. 82和1. 89,小孢子发育时期主要处于双核期,所占比例分别为62. 0%和75. 4%; V期、VI期、VII期花蕾的瓣萼比均大于2,花粉大多处于三核期,基本发育为成熟花粉,所占比例分别为63. 8%、85.3%和84. 5%。猕猴桃小孢子发育时期与花蕾形态特征存在密切关系,在猕猴桃单倍体育种取材中可依据花蕾形态特征采集适宜发育时期的小孢子细胞。In order to provide a suitable development stages of microspore for kiwifruit haploid breeding,this study used the buds of‘Donghong’kiwifruit as the material,which were stained with aceto-carmine dyeing solution and observed with biological digital microscope,and discussed the relationship between the bud morphological characteristics and pollen development stages.The results show that the ratio of petals to sepals length is 1.59 and 1.67 inⅠandⅡphase buds,and 84.4%and 68.6%pollens are in uninucleate stage,respectively;and the ratio of petals to sepals length are 1.82 and 1.89,respectively,and the pollens are mainly in binucleate stage(62%,75.4%)in III and IV phase buds.In V,VI and VII phage buds,ratio of petals to sepals length is all above 2,and the pollens are mainly in trinucleate stage,which develop into mature pollen,and the proportion is 63.8%,85.3%and 84.5%,respectively.In the study,the microspore development is closely related to the morphological characteristics of flower bud,so we can collect the suitable development stage of microspore cells according morphological characteristics of buds in kiwifruit haploid breeding.
