摘  要:目的探讨骨形态发生蛋白(BMP)复合多孔钛铌合金试件的体外成骨分化能力。方法将多孔钛铌合金试件分为单纯实验试件组(A组)和复合rhBMP-2实验试件组(B组),将两组试件分别加至兔骨髓间充质干细胞(BMSCs)中。通过倒置显微镜、扫描电镜、血清碱性磷酸酶(ALP)及骨钙素指标检测,观察BMSCs在两组钛铌合金试件中的生长情况及金属-组织界面的结合情况。结果 B组培养液中血清ALP浓度、骨钙素含量较A组明显提升;培养2周后电镜扫描示A组有大量细胞附着于材料外表面和孔洞内,细胞形态呈梭形,表现为单层生长,B组则表现为复层生长;培养4周后B组材料孔洞内可见钙沉积,而A组材料孔洞内未有钙沉积。结论多孔钛铌合金材料细胞相容性良好,兔BMSCs在试件中形态正常,具有良好的增殖分化能力;经BMP诱导,BMSCs在多孔钛铌合金材料中可向成骨细胞分化。Objective To study the osteogenic differentiation ability in vitro of bone morphogenetic protein(BMP)composited porous titanium-niobium(TI-NB)alloy specimens.Methods The porous TI-NB alloy specimens were divided into group A(no composited with BMP)and group B(combined with BMP),bone marrow mesenchymal stem cells(BMSCs)of rabbits were added into two groups.The growth of BMSCs in TI-NB alloy specimen and the bonding of metal-tissue interface in two groups were observed by inversion microscope,scanning electron microscope(SEM),and were analyzed by serum alkaline phosphatase(ALP)and osteocalcin examinations.Results The concentrations of serum ALP and osteocalcin in the culture solution in group B were significantly higher than those in group A.At 2 weeks after co-culture,plenty of cells were found in SEM attached to the outer surface and holes of the material with fusiform shape and single layer growth in group A;While in group B,the growth of compound layers was observed.After 4 weeks'co-culture,calcium deposition was found in the pores of porous TI-NB alloy material in group B,but there was no calcium deposition in group A.Conclusion Porous TI-NB alloy specimen is demonstrated to has good cell compatibility with rabbit BMSCs,which showing normal morphology and good proliferation,differentiation abilities;Induced by BMP,BMSCs in porous TI-NB alloy material can be differentiated into osteoblasts.
