摘 要:目的评估转铁蛋白-金纳米簇荧光分子探针的体内急性毒性。方法以转铁蛋白为模板,氯金酸为原料,合成转铁蛋白-金纳米簇荧光分子探针。利用透射电镜和粒度分析仪测定所合成纳米粒子的形貌、水合粒径;利用紫外-可见光和荧光分光光度计检测其光学特性。经小鼠尾静脉注射转铁蛋白-金纳米簇溶液,分时取血检测血常规、生化指标和急性时相反应蛋白。结果本研究制备的金纳米簇粒径为(1.8±0.2) nm,转铁蛋白-金纳米簇水合粒径为(48.7±8.6) nm,在波长280nm的有特征性蛋白吸收峰,最大荧光发射峰位于波长700nm处。白细胞数、急性时相反应蛋白在给药后会一过性升高,但均可在短时间内恢复至正常水平。结论采用模板法制备转铁蛋白-金纳米簇分子探针,方法简单高效,所制备的分子探针具有毒性低,生物相容性好等优点。Objective To evaluate the acute toxicity of transferring functionalized gold nanoclusters(Tf-AuNCs)in mice.Methods We adopted transferrin as a template to synthesize Tf-AuNCs.The as-prepared Tf-AuNCs samples were characterized by transmission electron microscopy(TEM),mastersizer,zeta potentiostat to detect the morphology,the hydrodynamic particle size.Fluorescence emission spectra and ultraviolet-visible absorption spectra were obtained on spectrafluorometer and UV/VIS spectrophotometer,respectively.Then,mice received one-time intravenous(i.v.)injections of Tf-AuNCs solution by the tail vein.The mice were sacrified and the blood was collected for acute phase protein,hematology,and biochemistry analysis at different time points.Results The Tf-AuNCs showed excellent chemical stability and optical properties.The average particle size of AuNCs was(1.8±0.2)nm and the average hydrodynamic size was(48.7±8.6)nm.The prepared Tf-AuNCs presented the emission peaks of fluorescence at the wavelength of 700 nm and the absorption peaks of Tf at the wavelength of 200 nm.The mice in the experiment group showed slight increase of white blood cell count(WBC)and the expression level of acute phase proteins of serum amyloid A(SAA)and C-reactive protein(CRP)at the early stage,but it restored to normal level soon.No abnormality was found in biochemical indicators.Conclusion The template approach has the features of simple preparation and high-efficiency.The Tf-AuNCs exhibit low toxicity and great biocompatibility.
