摘 要:Grassy Tiller1(GT1)属于植物HD-Zip Ⅰ亚家族转录因子,通过抑制侧芽萌动和侧枝伸长调控植物分蘖性状。本研究使用RT-PCR和RACE技术从甘蔗品种ROC22中克隆出甘蔗GT1同源基因ScGT1 (MK318528)。生物信息学分析发现该基因cDNA序列包含一个长度为723 bp的开放阅读框,可编码一个含240个氨基酸的蛋白。该蛋白具有一个Homeodomain保守结构域,分子量为26.44kD,理化等电点pI为6.04,属于亲水蛋白,无跨膜结构,不存在信号肽,可能定位于细胞核,参与氨基酸生物合成的可能性较高。蛋白结构及同源性分析表明该蛋白空间结构与高粱和玉米较为相似,Homeodomain结构域高度保守,变异较少。系统发育树分析表明该蛋白属于HD-Zip I亚家族中的gt1 Clade类群,与高粱和玉米GT1亲缘关系较近,初步推测ScGT1可能通过腋芽分生组织发育的调控来影响分蘖表现。该研究可为后续该基因更深入的功能鉴定和分子辅助育种奠定重要的前期基础。Grassy-tiller1 (GT1) is a transcription factor in the HD-Zip I subfamily,regulates the tillering trait by inhibiting the germination of axillary bud and the elongation of lateral branch.In this study,ScGT1(MK318528),a homolog of GT1 was cloned from sugarcane variety ROC22 by using the RT-PCR and rapid amplification of cDNA ends(RACE) techniques.Bioinformatics analysis showed that the cDNA sequence of ScGT1 contain a complete open reading frame (ORF) with a length of 723 bp which can encode a protein of 240 amino acids.ScGT1 protein has a conservative homeodomain with a molecular mass of 26.44 kD and bioelectric point value of 6.04.This protein belongs to hydrophilic protein,has no transmembrane structure and signal peptide,which is probably located in the nucleus and participates in amino acid biosynthesis.The comparing analysis in structure and homology of protein among gramineous plants indicated that ScGT1 shared high similarity with the homologs of Sorghum bicolor and Zea mays in protein structure,especially in the homeodomain with less variation.The phylogenetic tree among these GT1 homology proteins from different gramineous plants demonstrated that ScGT1 belonged to the gt1 Clade cluster in HD-Zip I subfamily,and had a close relationship with the GT1 proteins from Sorghum bicolor and Zea mays.ScGT1 may regulate the axillary meristem in the development of tillers was a preliminary speculation in this study.The results provided a theoretical foundation for function analysis of ScGT1 and molecular aid selection of sugarcane yield in the future.
