摘 要:为了解细鳞斜颌鲴(Xenocypris microlepis)的生化遗传特征,并以此为基础为其种质资源鉴定提供理论依据,本研究采用不连续聚丙烯酰胺凝胶垂直板电泳对细鳞斜颌鲴5种组织(心脏、晶状体、肌肉、肾脏和肝脏)的乳酸脱氢酶(LDH)和苹果酸脱氢酶(MDH)进行分析。结果表明:所测样本鱼的心脏、晶状体、肌肉、肾脏和肝脏中的LDH酶带数分别为:6~8、5、2~4、5和4~6条; MDH酶带数分别为:7~8、5~6、7~8、3~4和4条;细鳞斜颌鲴的LDH同工酶和MDH同工酶具有组织特异性,其心脏、肾脏和肝脏MDH分为上清液型(s-MDH)和线粒体型(m-MDH),肌肉和晶状体组织中未见上清液型MDH(s-MDH)。研究认为晶状体组织LDH可作为鉴定细鳞斜颌鯝种质特征的生化遗传参数。本研究可为鉴定细鳞斜颌鲴种质、制定其种质标准提供参考依据。This paper was to study the morphological and biochemical genetics characteristics of Xenocypris microlepis so as to provide theoretical basis for their germplasm resources identification. By using the vertical polyacrylamide gel electrophoresis, lactate dehydrogenase (LDH) and malate dehydrogenase (MDH) isozymes from five tissues (heart, eye, muscle, kidney and liver) were analyzed . The results showed that: the band numbers of LDH isozymes expressed in heart, eye, muscle, kidney and liver were 6-8, 5, 2-4, 5 and 4-6, respectively, and that of MDH were 7-8, 5-6, 7-8, 3-4 and 4, respectively. These two kinds of isozymes from Xenocypris microlepis presented tissue specificity. Briefly, MDH isozymes expressed in heart, kidney and liver of Xenocypris microlepis were divided into supernatant type (s-MDH) and mitochondria type (m-MDH). The supernatant type of malate dehydrogenase (MDH) isozymes could not be detected in muscle and eyes. And lactate dehydrogenase (LDH) isozymes expressed in eyes can be used as the biochemical genetic marker of Xenocypris microlepis. This research provided a reference basis for the establishment of the germplasm standards and the germplasm identification of Xenocypris microlepis.[Chinese Fishery Quality and Standards,2019,9(1):10-17].
