摘  要:目的筛选角质形成细胞应对红色毛癣菌感染的差异表达基因与关键基因。方法构建红色毛癣菌感染人工表皮模型与阴性对照组一起进行RNA-seq测序分析。结果在红色毛癣菌孢子感染人工表皮后共筛选出了1 564个差异表达基因,包括1 102个上调基因与462个下调基因。这些差异基因的Go分析结果主要富集于生物学调控、膜及膜组分和催化功能。通过kegg通路分析,差异表达基因主要富集于与结直肠癌、赖氨酸降解和肌醇磷酸代谢相关通路。另外,蛋白互作分析发现PHLPP2和TOP2B这2个基因节点的度最高,可能在人角质形成细胞应对红色毛癣菌感染中具有重要的作用。结论筛选出了1 564个角质形成细胞应对红色毛癣菌感染的差异表达基因,这些基因可能在人角质形成细胞应对红色毛癣菌感染中具有重要作用。Objective To screen the differentially-expressed and key genes in the keratinocytes in response to Trichophyton rubrum infection. Methods The artificial epidermis model infected by Trichophyton rubrum was established. RNA-seq was performed in the reconstructed human epidermis and negative controls. Results A total of 1 564 differentially-expressed genes were identified, including 1 102 up-regulated genes and 462 down-regulated genes after the artifical epidermis was infected by Trichophyton rubrum. The gene ontology (GO) analysis indicated that the majority of the target genes were related to biological regulation, membrane, membrane component and catalytic function. Kegg pathway analysis revealed that most target genes were distributed in the colorectal cancer, lysine degradation and inositol phosphate metabolism signaling pathways. In addition, protein-protein interaction networks demonstrated that PHLPP2 and TOP2B had the highest degree of connected node, which might play a critical role in keratinocytes challenged with Trichophyton rubrum. Conclusions A total of 1 564 differentially-expressed genes are screened in the keratinocytes in response to Trichophyton rubrum infection, which probably play a pivotal role in keratinocytes against Trichophyton rubrum infection.
